Abstract


Objective To determine the protective effect of Sodium Salicylate against damages of the NIT-1 cells induced by IL-1βand the possible mechanism.Methods The pancreaticβcell line NIT-1 cells was provided by . The cells untreated were enrolled as controls; the cells treated with IL-1β(5ng/ml) were enrolled as impaired group; and the cells treated with IL-1β(5ng/ml) and Sodium Salicylate(5mmol/l, 10mmol/l, 20mmol/l) were enrolled as protected group. IN separate cultures, apoptosis was measured by flow cytometry with propidium iodide staining and 24h was selected as the termination point. The expression of NF-KB in nuclear was measured by western blot analysis and the termination points include 0h, 1h, 12h, 24h.Results (1) The apoptosis cell fractions of the group treated by IL-1βalone was significantly higher than that of untreated group.(2) The apoptosis cell fractions of the group treated by IL-1βand Sodium Salicylate (10mmol/l or 20mmol/l) was significantly lower than that of the group treated by IL-1βalone.(3) The expression of NF-κB of the group treated by IL-1βalone was significantly higher than that of untreated group.(4) The expression of NF-κB of the group treated by IL-1βand Sodium Salicylate (10mmol/l or 20mmol/l) was significantly lower than that of the group treated by IL-1βalone.(5) The expression of NF-κB of the group treated by IL-1βand Sodium Salicylate (20mmol/L) is more or less than that or untreated group at any observed time. Conclusions (1) IL-1βmay induce the apoptosis of NIT-1 cells.(2) IL-1βmay activate NF-κB.(3) Sodium Salicylate inhibits IL-1β-induced apoptosis.(4) Sodium Salicylate inhibits IL-1β-induced activation.(5) The effect of IL-1β-induced apoptosis can be inhibited, when the concentration of Sodium Salicylate reaches up to 20mmol/L.
