Abstract


Ginsenoside -Rd (Ginsenoside-Rd, GR-d) is a monomeric compounds extracted from precious Chinese medicine Notoginseng is a novel neuroprotective drugs . Recent studies have found that it has specific blocking the function of the cell membrane receptor-dependent calcium channels , inhibition of glutamate release , also has multiple roles in antioxidant protection mitochondria . The occurrence and modulation of glutamate and NMDA receptors and neuropathic pain (neuropathic pain, NPP) is closely related , suggesting that the GR-d may be a potential NPP analgesics . The subject of its NPP whether the analgesic effect and its related mechanisms were discussed. In this study, first make SD rat sciatic nerve selective injury (spared nerve injury, SNI) pain model and detect mechanical stimuli ( paw withdraw mechanical threshoid the flinching threshold PWMT ) before and after the administration changes observed GR-d SNI caused allodynia (allodynia) effect ; Secondly, the GR-d in the corresponding segments of the spinal cord of glutamic acid , and NR2A? B expression observed using immunofluorescence staining method . Found that SNI postoperative 10 d the surgical side PWMT values ??significantly lower than the control group and the sham group , 20 d after the lowest value , the the SNI Saline group was significantly higher than in the SNI G-Rd group PWMT value ; immunofluorescence staining SNI postoperative 20 d, spinal cord L4-6 segments the surgery paranotum horn against glutamate -like and anti- NR2A / B immunoreactivity in the average fluorescence intensity of the product (the mean fluorescent intensity, MFI) was significantly higher than the control group spinal cord segments L4-6 the surgery paranotum horn of anti glutamate - like immunoreactive products MFI ; SNI G-Rd group and the sham group was significantly lower than the SNI and SNI Saline group , while the anti- NR2A / B MFI no significant changes . In summary , this research shows , GR-d can significantly improve the SNI rats the hyperalgesic behavior , one of the possible mechanisms of spinal cord reduce its effective content related to corresponding segments of the spinal cord dorsal horn glutamate . These results laid the theoretical foundation for clinical analgesic application for GR-d .
