Preparation of Borneol and Brucea Javanica Oil Nanoemulsion and Study on Its Inhibitory Effects on Proliferation of Brain Glioma in Rats

Abstract


Objective To prepare borneol and Brucea javanica oil nanoemulsion, to inspect its physical and chemical properties, stability experiment and construction of C6rat glioma models, and the in vivo rat glioma tumor suppression effect is studied. In order to improve the inhibition of glioma of Brucea javanica oil and improve the therapeutic effect by using nanoemulsion as the carrier and combined with borneol.Methods The content of oleic acid in Brucea javanica oil was determined by GC after pre-column derivatization. The borneol and Brucea javanica oil nanoemulsion were prepared by the steering method. To draw the pseudo-ternary phase diagram with titration method, screen suitable emulsifier and auxiliary emulsifier. Examined emulsifier, auxiliary emulsifier, oil phase and Km in different system which effect on nanoemulsion area, ultimately determined a set of best prescription by the phase behavior of the various systems and the comparison of the nanoemulsion regional area. Morphology of borneol and Brucea javanica oil nanoemulsion were observed by transmission electronmicroscope. Mean particle size was estimated by laser particle size analyzer. The content of oleic acid were detected by high efficiency gas chromatography. The stability of bomeol and Brucea javanica oil nanoemulsion were examined. The inhibition of rat tumor, the tumor change in weight and volume with model group, Brucea javanica oil injection group, Brucea javanica oil nanoemulsion group and borneol and Brucea javanica oil nanoemulsion group were investigated by building a C6rat glioma model.Results Established the GC analysis method for determination of oleic acid content in the Brucea javanica oil, and there is a good linear relationship between0.15~0.75mg/mL (r=0.9999). Borneol and Brucea javanica oil nanoemulsion prepared by the optimum formulation of IPM35%, Brucea javanica oil35%(1%borneol), Cremopher RH4018%, Labrasol12%. The appearance of nanoemulsion is spherical; the mean particle size was (47.60±0.57) nm; The PDI was (0.22±0.01); Zata potential was (-1.06±0.12) mV; The content of oleic acid was (0.4241±0.0056) mg/mL, and nanoemulsion stability is good. The rat C6glioma model was successfully constructed. Antitumor effect in vivo experiments showed that the tumor weight of model group, the Brucea javanica oil injection group, the Brucea javanica oil nanoemulsion and borneol and Brucea javanica oil nanoemulsion group were respectively(1.32±0.19)g,(0.84±0.08)g,(0.76±0.06)g,(0.57±0.10)g; The tumor inhibition rate about tumor weight of the Brucea javanica oil injection group, the Brucea javanica oil nanoemulsion and borneol and Brucea javanica oil nanoemulsion group were respectively36.49%,42.80%,56.94%; While the tumor volume of model group, the Brucea javanica oil injection group, the Brucea javanica oil nanoemulsion and borneol and Brucea javanica oil nanoemulsion group were respectively (72.2±9.4)mm3,(44.2±5.1)mm3,(42.3±4.9)mm3,(27.9±2.5)mm3; The tumor inhibition rate about tumor volume of the Brucea javanica oil injection group, the Brucea javanica oil nanoemulsion and borneol and Brucea javanica oil nanoemulsion group were respectively38.8%,41.4%,61.3%. Brucea javanica oil can effectively inhibit the growth of glioma cells, and even borneol and Brucea javanica oil nanoemulsion enhanced inhibition of rat glioma.Conclusion The surface of borneol and Brucea javanica oil nanoemulsion prepared by steering method was smooth and round, the particle size distribution was symmetrical, and nanoemulsion had high content of oleic acid and good stability. Antitumor effect in vivo experiments showed that borneol and Brucea javanica oil nanoemulsion has a strong inhibitory on rat glioma, improve the efficacy of Brucea javanica oil for the treatment of brain glioma, which is conducive to development Brucea javanica oil as a treatment for glioma drug market prospects. The results showed important theoretical and practical reference value for design, development and evaluation of of anticancer drugs targeting agents.
