Abstract


Tissue survival is the key procedure on a surgical operation. The main cause oftissue necrosis is induced by ischemia/reperfusion (I/R) injury. The mechanisms ofI/R injury include hypoxia, inflammation, and free radical damage. So, for a long time,researchers involved in finding a proper way of prevent I/R injury. Hydrogen gas hasbeen reported to alleviate cerebral I/R injury as a free radical scavenger. Theprotective effect of hydrogen-rich saline showed a promising perspective.Here, Rat skin flap model were constructed for the research ofIschemia/reperfusion injury. The flap samples were harvested from the proximal,middle, and distal part of the viable flap and cytokines expression were detectedseparately in order to assess whether there were significant differences among thethree parts and confirm an equitable way of sampling on ischemia/reperfusion injuryof rat skin flap model. At the same time, the model were use to evaluate the repairfunction of saturated hydrogen saline in the flap ischemia-reperfusion injury.40male SD rats with the same growth condition were divided into3groupsrandomly, Sham operated group, Ischemia/Reperfusion group, andIschemia/Reperfusion with hydrogen-rich saline injected group. The area of viableflaps were measured, and samples were harvested on postoperative day5. The level ofIL-1β, IL-6, TNF-α, EPO and MDA were detected to assess the sampling way.Statistical analyses were performed using SPSS.The flap survival rate of IR group was just1/5of sham group. The flap survivalrate of IR/H group was raised2times compared with IR group. Significantdifference in cytokines levels were noted between the Sham groups and IR group.Significant difference in cytokines levels between the IR groups and IR/H group wasalso noted. But no statistical difference in cytokines levels in either inter-group.A certain concentration of saturated hydrogen saline has curative effect obviouslyto the flap ischemia-reperfusion injury. Samples can be harvested at any arbitrarypoint on a skin flap to stand for the inflammatory level of the whole flap. And itshould be sampled from the same position on different flaps in the same experiment.However harvest from the parallel point were encouraged for taking parallel samples.
