ObjectiveTo construct the model of rat’s pulmonary contusion, determine theexpression of aquaporin1(AQP1) and probe its relationship with pulmonaryedema and investigate the role of AQP1in rat’s pulmonary contusion.Methods40SD rats which were healthy and grow-up and weighed2005gram werechoosed. They were randomly divided into experimental and controlgroups,which group had20SD rats. The model of rat’s pulmonary contusionwas estalished in the experimental group. There were some prerequisites tosucceed: the metal object weighs180gram, the hitting area which’s acreagewas2.83cm2is round, the hitting height is1meter. The ratio of wet to dry lungweight was measured, and the changes of lung tissue were pathologicallyobserved. The expression and distribution of AQP1in lung tissue wereexamined by immunohistochemical method.ResultsThere were serveral characteristics of lung tissue in the experimental group:pulmonary interstitial edema and pulmonary alveolar edema in varying degrees,the broader alveoarcapillary membrane, a large number of erythrocytes inalveolar spaces, the damaged structure in part of alveolar spaces andinflammatorycell infiltration in pulmonary interstitial. The ratio of wet to dry lungweight increased significantly (P<0.01), meanwhile the expression of AQP1inthe experimental group decreased significantly compared with the control group(P<0.01).Conclusions1.The reliable and stable model of SD rats’ pulmonary contusion was established by using free-falling metal object to hit rats’ right thorax.2.The decreasing expression of AQP1in the experimental group probablyprevents lung tissue reabsorbing fluid of pulmonary interstitial and transportingthe fluid between alveolar space and pulmonary capillary, which may cause oraggravate pulmonary dysfunction.
