Abstract


ObjectivesUse of RNAi to silence gene expression of CCR3in eosinophils, blocking thepath of Eotaxin/CCR3to konw the changes of eosinophils in the bone marrow,peripheral blood, nasal lavage fluid and local tissue, and observe its impact oneosinophils apoptosis process, in order to understanding the role and impact ofeosinophils in this process, find new treatments for the complete cure of allergicrhinitis.MethodsCCR3siRNA plasmid(pLVX-ShRNA2-mCCR3-1+2+3+4) effective for andspecific to the inhibition of CCR3gene expression obtained from in vitroexperiments was used in the murine allergic rhinitis via nasal administration ofliposomes, with HE staining and Wright stain to detect the changes of eosinophils inthe nasal mucosa of mice, blood, bone marrow, nasal lavage fluid, separately.Results①After HE staining, a microscope was used to reveal that the swelling of thenasal mucosa, the detachment of epithelia, and the increase of the infiltration ofinflammatory cells and amount of eosinophils in model group compared with controlgroup, while RNAi treatment decreased the amount of eosinophils and the swellingof the nasal mucosa.②After wright staining, by counting the typical eosinophilsproportion in the blood, bone marrow, nasal lavage fluid were higher than normalcontrol group, after RNAi treatment, it is decline in the proportion.ConclusionThis study suggests a therapeutic approach the genes CCR3, through intranasalmucosal administration of target gene siRNA, can alleviate the animal model withallergic rhinitis and reduce eosinophilic quantity of bone marrow, peripheral bloodand nasal lavage fluid.
