Abstract


Dopamine can form polydopamine coatings through Self-polymerization. The polydopamine coatings were rich with active groups. Modification functional molecules such as anticoagulant molecules or promoting cell adhesion and cell proliferation is realized by the polydopamine coatings. To covalently immobilize various biomolecules such as promoting the endothelialization, anti-proliferative, anti-inflammatory or anti-bacterial etc onto same surface, which realize surface multi Functionalizations, we construct a film and it can meet the requirements:first, it has a good specific biological function (such as anticoagulant property); second, it still keeps the secondary reaction activity of graft the biomolecules of traditional polydopamine coatings.The research has not been reported before.Firstly the research prepared dopamine-heparin complex solution by mixing dopamine and heparin solution, then adjusted pH, immobilized heparin onto titanium surface through dopamine Self-polymerization.Graft Arg-Gly-Asp (RGD) peptide on dopamine-heparin anti-coagulation films and study graft method to realize balance of anti-coagulation and promoting endothelialization.First, The dopamine-heparin complex films were fabricated via set different concentrations of dopamine and heparin and different deposition time.The process were optimized via test of heparin content and activity, surface hydrophilicity, platelet adhesion and Activated Partial Thromboplastin Time (APTT), etc.Then, different concentrations RGD peptide on dopamine-heparin anti-coagulation films were grafted. Best concentration of graft RGD peptide was determined via platelet adhesion and activity, cells culture.The samples were characterized by Fourier Transform infrared spectroscopy (FTIR) X-ray Photoelectron Spectroscopy (XPS), immunofluorescence staining, water contact angle and Enzyme-Linked Immuno Sorbent Assay (ELISA), etc. The results show that heparin was immobilized onto sample surfaces and still keeps the high activity. RGD peptide was graft onto the dopamine-heparin complex films. The film still keeps the secondary reaction activity of traditional polydopamine coatings was preliminarily confirmed through covalent graft RGD peptide.Hemolysis rate test, coagulation time, platelet adhesion and blood test results show that after fabrication dopamine-heparin complex films on Ti.
