Abstract


Chinese mitten crab (Eriocheir sinensis) is a special aquatic economicalorganism in China and plays a special important role in Chinese aquacultureindustry. In this study, combining culture biology, molecular biology andmolecular genetics and field investigation, outdoor experiment and indoor,The effects of formulated diet、fresh fish on growth, fatty acid compositionand gene expression in hepatopancreas of juvenile crab were analyzed.Meanwhile, genetic difference and relationship were investigated between theeven and odd year populations of Chinese mitten crab from the Yangtze River.We hope these results could support theoretical and technical helps to pushthe development of aquaculture and resource utility of Chinese mitten crab.Part1. The impact of formulated diet、fresh fish on growth and fatty acidcomposition in hepatopancreas of juvenile crab.Formulated diet and iced small fish have been widely used as the most important feed in Chinese mitten crab aquaculture industry. In this part, theimpact were evaluated of formulated diet and fresh fish (tilapia) on growthand fatty acid composition in hepatopancreas of Chinese mitten carp atjuvenile stage, which were hold in nets fixing in the same pond for130days.The results showed that there were not a significant differencein weight, shelllength, specific growth rate (SGR) and hepatopancreas index (P>0.05)between the formulated diet and fresh fish (tilapia). However, the juvenilecrab fed with tilapia displayed significantly high survival rate (P<0.05) thanthose fed with formulated diet, and significantly low precocious rate andweight gain rate than formulated diet. The main fatty acid in hepatopancreasincluded C18:1n-9、C16:0、C18:2n-6、C16:1n-7, but the content of n-6PUFAwas significantly higher than n-3PUFA. There were significantly highercontents of C16:1n-7、C14:1n-5、C17:1n-7、C22:1n-9、C24:1n-9、C18:3n-3、C20:5n-3、C22:6n-3in crab fed with formulated diet than those fed withtilapia(P<0.05), but significantly lower contents of C18:1n-7、C18:1n-9、C20:1n-9、C18:2n-6in crab fed with formulated diet compared to tilapia(P<0.05). Moreover, male crab showed slightly higher contents of C18:2n-6、C18:3n-3、C20:5n-3、C22:6n-3than female crab in both formula diets andfresh tilapia with no significant level (P>0.05).Part2. The impact of formulated diet、fresh fish on the mRNA expressionof FAS、FABP、ALF genes in hepatopancreas of juvenile crab The experiment was conducted to investigate the effect of formulateddiet、fresh fish on the fatty acid mixture, transport, anti bacterium inhepatopancreas of juvenile crab. The expression differences of three genes,fatty acid synthase (FAS), fatty acid binding protein (FABP) andanti-lipopolysaccharide factor (ALF), were analyzed from juvenile crab fedwith formulated diet and tilapia for130days. The results indicated:①themRNA expression of FAS fed with tilapia were significantly higher than thosefed with formulated diet (P<0.05);②the mRNA expression of FABP fed withformulated diet were significantly higher than those fed with tilapia;③themRNA expression of ALF fed with tilapia were higher than those fed withformulated diet, but there was no significantly difference between them(P>0.05);④there was no significantly difference between the female and malecrabs that fed with the same diet. The results of this study demonstrated thatformulated diet、fresh fish can cause significantly difference about the relativemRNA expression in hepatopancreas of juvenile crab.Part3. Genetic differentiation analysis of the even and odd yearpopulations of Chinese mitten crabIt is a putative hypothesis that there were two lineages or categories ofthe odd and even years for Chinese mitten crab. In the study, for testing this hypothesis, genetic differentiation was analyzed for eight wild populations(sampled in2004、2006~2011) from the Yangtze River based on18microsatellite loci. The results indicated that:①the HOand HEof the oddyear populations were significantly lower than that of the even yearpopulations (P <0.05);②There were significant pairwise FSTdifference(P=0.024) between the odd and even year populations;③The phylogeneticdendrograms based on NJ methods showed obvious two clades among them,one clade consisted of the even year populations, one clade consisted of theodd populations;④Both Principal coordinate analysis and STRUCTUREgenetic clustering analysis indicated that significant genetic differentiationbetween the odd and even year populations, but slight genetic introgressionwould had occurred between them;⑤Genetic bottleneck signal was detectedin the two even year populations (2004,2006) and odd year populations (2007,2011). The results of this study demonstrated that the Chinese mitten crab inYangtze River exist two lineages of the odd and even year populations,however there were a level of gene glow between the two lineages.
