The Induction of M2Polarization of Kupffer Cells in Rats with Severe Acute Pancreatitis in Vitro

Abstract


Objective:To explore and compare the role of IL-4and Treg on the induction of M2polarization of kupffer cells in rats with severe acute pancreatitis(SAP) in vitro.Methods: The SAP rat model was established by retrograde pancreatic ductal injectionof5%sodium taurocholate. Kupffer cells were isolated from sham opration rats and ratswith acute pancreatitis. As to rats with acute pancreatitis, Kupffer cells were isolated at24hours after induction of AP and there were3groups with different treatment in Kupffercells from rats with AP:PBS、 IL-4and Treg. Furthermore,3time point were set in eachgroup:4h,8h and16h. Spleen Tregs were extracted by CD4+CD25+immune magneticbead sorting method, and these Tregs were used to intervene in kuppffer cells incorresponding groups. The expression of IL-1β,IL-10were detected by reversetranscription polymerase chain reaction(RT-PCR) and immunofluorescence confocal.Results:(1)Compared with SO group, the expression of IL-1β was distinctively increasedin each SAP+PBS group(P<0.01).(2) Compared with SAP+PBS(8h) group, theexpressions of IL-10was increased in SAP+IL-4(8h) group(P<0.05),but markedlyincreased in SAP+Treg(8h) group(P<0.01). Compared with SAP+PBS(8h) group,theexpressions of IL-1β was decreased in both in SAP+IL-4(8h) group and SAP+Treg(8h)group.(3)At the same time point, the expression of IL-10in SAP+Treg group wasmarkedly higher than in SAP+IL-4group(P<0.01).Compared with SAP+PBS(4h) group,the expression of IL-10in SAP+IL-4(4h) was no obvious deferences,However, theexpression of IL-10in SAP+Treg(4h) group was markedly higher (P<0.01).(4)In thisexperiment,the expression of IL-10in SAP+IL-4groups had no significant changes overtime, but the expression of IL-10in SAP+Treg groups firstly descended and then rose. Conclusions:After the occurrence of SAP, the M1polarization of kupffer cells release lotsof proinflammatory factors(such as IL-1).Both IL-4and Treg can induce the M2polarization of kupffer cells, M2polarization kupffer cells produce a large number ofanti-inflammatory factors(such as IL-10). Compared with IL-4, the role of Treg on theinduction of M2polarization of kupffer cells is more prompt and effective.
