The Application of the Nano-leveled Attapulgite in Mammals Cell Cultivation

Abstract


Attapulgite (ATP), which is a crystalline hydrated magnesium aluminum silicate with unique three-dimensional nano-structure and fibrous morphology, was discovered in the 19th century. Due to its structural morphology, the Attapulgite has a wide variety of industrial application, but there is only a little report on its application in the mammalian cell cultivation.In this work, the cell line Sp2/0 (the cell Mouse myeloma) and Vero cell (the cell African monkey kidney cell) were employed to research the Attapulgite effect on the mammalian cell cultivation by measuring the cell viability, the cell growth curve, the nourishing ingredient in medium and the change of the metabolite, respectively. In addition, we initially explore the mechanism of the ATP effect. Main contents of this work are as follow:(1) Purify the Attapulgite. Three methods are adopted in this paper, the Sodium Polyacrylate, Hydrochloric acid and ultrasonic used for purified the Attapulgite. Then the SEM, TEM were used to observe the photograph of the purified Attapulgite. It was concluded that acidification Attapulgite is the best method for cell cultivation. Because the Attapulgite’surface area is increased and the Attapulgite is purified.(2) Sp2/0 and Vero cell cultivation with ATP regulation. The cell viability, the cell growth curve, the nourishing ingredient in medium and the metabolite are measured to assess the ATP positive effect on the mammalian cell cultivation, respectively. The results show that the ATP can optimize the cell cultivation environment and improve the cell cultivation efficiency.(3) Mechanism of the ATP effect on mammalian cell cultivation. We employed the FT-IR spectra, XPS and XRD to characterize the physical and chemical properties of the ATP fore-and-aft the Mammals cell cultivation, and initially explored the mediated-mechanism of ATP for mammalian cell cultivation.
