The Effect of Dietary Energy Level on Ovarian and Uterine Expression of LHR and FSHR mRNA in Prepubertal Gilts

Abstract


The main objective of the present study was to examine the ovarian and uterineexpression of LH receptor (LHR) and FSH receptor (FSHR) mRNA, under long-termtreatment with different levels of dietary energy intake, in prepubertal gilts. Ninegrowth-matched littermate hybrid gilts(Landrace × Large White × Duroc) were used inthe present study. At 50d of age and 30 kg BW, gilts had ad libitum access to feed for90 days(d1 to d90) . From d91 to d112, littermates were divided and allotted into oneof the three treatments, fed the diet of high energy level(n=3, 120% of NRC energyrequirements), moderate energy level(n=3, 100% of NRC energy requirements), orlow energy level(n=3, 80% of NRC energy requirements). A free access to water wasprovided throughout the whole research period. Ovaries and uteruses were collected,at the slaughter after 3 weeks energy treatment, and processed for determination of therelative quantities of LHR and FSHR mRNA using semi-quantitative RT-PCR. On theovaries the high compared with the moderate and the low dietary energy level resultedin a significantly greater amount of LHR and FSHR mRNA(p<0.01), and the lowcompared with the moderate also showed a significantly fewer amount of LHR andFSHR mRNA(p<0.05). While in the uteruses, there were no significant differences canbe detected, among the amount of uterine LHR and FSHR mRNA from the gilts underlong-term treatment of different dietary energy intake levels, by the use ofsemi-quantitative RT-PCR. In the present study, high energy intake level markedlyenhanced the ovarian expression of LHR and FSHR in prepubertal gilts, whereas, lowenergy intake level markedly inhibited such expressions. While in the uterus, therewere no significant differences can be detected, of the uterine LHR and FSHRexpression at the mRNA level, among the gilts under long-term treatment of differentdietary energy intake levels, by the use of semi-quantitative RT-PCR. And that showedremarkable significance in understanding and explaining the molecular mechanism ofenergy influencing reproductive performance in prepubertal gilts.
