The Effect of Progesterone on the Expression of the β-defensin in Ovine Oviductal Epithelial Cells

The method of isolation and primary cell culture of ovine oviductal epithelial cells was studied in this research, and the effect of the different concentrations of progesterone on expression ofβ-defensin in ovine oviductal epithelial cells was also investigated in this paper. The results indicated that the biological characteristics of epithelioid cells unchanged in culture medium (Dulbecco’s Modified Eagle’s and HAM’S F-12, containing 20% standard FBS, penicillin andstreptomycin).Immunohistochemical stain with keratin showed positive. The results from semiquantitative RT-PCR by supplementing with different concentrations of progesterone in ovine oviductal epithelial cells with serum free medium revealed that the effect of the progesterone on expression ofβ-defensin were significant. The expression ofβ-defensin treated with progesterone groups were significant higher than controls(P<0.05). Total expression ofβ-actin treated with progesterone was not significant. There was no difference in supplementing with 10-7,10-8 , 10-9 mol/l progesterone, but the expression ofβ-defensin treated with 10-8 mol/l progesterone was highest in three groups.
