With more and more studies of diabetes and obesity, a strong correlation between type 2 diabetes and obesity has been recognized. One of the common characters of obesity and type 2 diabetes is chronic elevation of circulating free fatty acids (FFAs). Recently it has been suggested that PTEN plays an important role in insulin metabolism, but there is no report about whether PTEN participates in the impairment of pancreatic p cell induced by FFAs; Recently, uncoupling protein 2 (UCP2) and mitochondrion and FFAs were observed to be related with the defects of pancreatic p cell, but the mechanism is not clear.Objective:To investigate the effects of free fatty acids (FFAs) on the expressions of PTEN and uncoupling protein 2 (UCP2) and mitochondrial membrane potential in pancreatic PTC3 cell line, and to explore the mechanism of pancreatic p cell impairment induced by FFAs.
