Effect of Conjugated Linoleic Acid on Gene Expression of Lipogenesis and Lipolysis in Liver of Mouse

Conjugated linoleic acid (CLA) which is unsaturated fatty acid includes conjugated double bonds and has many kinds of geometrical isomers and positional isomers. After being researched many years, CLA is discovered that it has effects of reducing body fa、losing weigh、anticancer treatment、anti-atherosclerosis、enhancing immunity、improving insulin resistance and so on. Because of CLA having so many kinds of important physiological functions, it already has been honored as "the 21st century’s new nutrient". Although CLA are caused on by researchers from various countries, its regulating mechanism are still not very clearly. This study includes following works:1. Twenty-four femal KunMing rats were randomly designed as control group and dietary with 1% CLA group. The rats were weighted and the body weight gain were recorded. The results showed that body weight gain of rats were significantly reduced by supplementation with CLA. The experiment indicated that dietary CLA could enhance adipose metabolism.2. Blood of the rats was gathered and then the glucose (GLU) level, the total cholesterol (CHO) level, the triglycerides (TG) level, the low density lipoprotein (LDL) and the high density lipoprotein (HDL) level of control group and experimental group were analyzed. The results showed that supplementation with 1% CLA reduced LDL、CHO、TG、and GLU level obviously (P< 0.01); the HDL level was higher than the control group(P<0.01). The experiment indicated that CLA could promote adipose metabolism of KunMing rats.3. The total RNA was extracted by the TRIzol Reagent from livers of rats. cDNA was synthesised after RT-PCR. Primers were designed and semi-quantitative PCR assay was carried on, then the results were analysised by gel documentation system and Bandscan software. The results suggested that GAPDH expresstion of the experiment group was same as the control group’s; PPARy and FAS expresstion of the experiment group were obviously lower than the control group’s; HSL and TGH expression of the experiment group were obviously higher than the control group, the difference was significance (P<0.01). The results indicated that CLA could promote adipose metabolism, and modulate lipid metabolism by regulating genes relating to its metabolism. This research provided the foundation for the following researchers and evidence for studying regulating mechanism of CLA in fat metabolism in the future.
