Studies on Screening of Nitrogen Nutrition Mutant and Its Nitrogen Nutrition Mechanism

Abstract


In this paper, nutrient solution culture methods to seedling apparent symptoms of reference indicators produced by Agrobacterium T1 generation mutants were screened , and the mutant phenotype nitrogen nutrition pedigrees molecular detection of mutations were isolated Nitrogen pedigree further from nitrogen nutrition characteristics and root characteristics were validation studies . The main results are as follows: one through the flowers 11T-DNA insertion mutants of 2173 T1 generation pedigrees nutrient solution culture screening of mutations observed in pedigree 152 , accounting for 6.99% screening population . Mutant types are divided into five categories, namely nitrogen auxotrophic mutant , mutant roots , albino type , etiolated seedlings type dwarf plant type, the mutation rate was 1.24% , 0.33% , 4.23% , 0.87% , 0.33 percent . 2 PCR testing showed that mutations in 27 pedigrees Nitrogen has 14 families were separated . Nitrogen expressed as mutant pedigree : small plant, the leaves are small , yellowish green , hair roots slow, poor fine roots and root hairs , root more yellow , white and poor root development . 3 through 12 consists of home -based T1 -and T2 generation of different nitrogen concentrations culture results showed that 955 lines at different nitrogen levels T1 -and T2 represents now the same separation characteristics , indicating that the mutation and T-DNA insertion directly related , through genetic analysis and inspection , found that mutations in the T1 generation pedigree 955 T2 generation segregation ratio of 3:1 are the inheritance of 955 tied with a mutated gene and showed recessive mutations. 4 through 955 T2 generation plants root system characteristics and differences in nitrogen metabolism studies have shown that : 955 line mutations in plants as ammonium nitrate absorption and uptake rates were reduced , the plants to reduce nitrogen content , nitrate reductase activity decreased, plant nitrogen assimilation capacity decreased, thus affecting root growth and development , the performance of shorter roots , root volume and root active area smaller , plant height and chlorophyll SPAD value decreases.
