Abstract


Vitamin B12 is one of the important microbial fermentation products, widely used in medicine, food and livestock production by microbial fermentation. The papers from the application point of view, the initial substrate concentration, acid on cell growth and the synthesis of cobalamin, exploring the line separating propionic acid in fermentation engineering, the lifting of the organic acids (propionic acid) on the growth of bacterial cells suppression, expect to provide a reference for cobalamin fermentation production process improvements. This thesis is mainly the following aspects of the study. . Of substrate concentration on the influence of vitamin B12 fermentation investigated the concentration of the C source and N source concentration of Propionibacterium freudenreichii affect fermentative production of vitamin B12, the experimental results show that the glucose concentration was maintained at 2.0%, corn steep liquor concentration is maintained at 4.0% When most conducive to microbial growth. Taking into account the operability of the feeding during the fermentation process, the glucose concentration was maintained at 1.0% -3.0%, the concentration of the corn syrup is maintained at 3.0% -4.0% is more reasonable. Propionic acid fermentation of vitamin B12 in this experimental study plus propionate Propionibacterium freudenreichii growth effects of propionic acid suppression with replacement of fresh medium lift the Propionibacterium freudenreichii fermentation production of vitamin B12 the impact. The study showed that the concentration of propionic acid is applied at 0-0.5% bacterial μmax reduced to 60%, plus propionate 1.0% bacterial μmax to 30%. Replaced with fresh medium lift propionic acid suppression, biomass increased by 74.5%; deoxyadenosine cobalamin production increased by 40.7%.
