Abstract


Watermelon variety, rich flesh color , red , pink , orange, yellow , white and other , mainly different varieties of watermelon flesh -colored carotenoids type and content due to a difference . Analysis and detection of watermelon watermelon carotenoids for processing and to provide the basis for watermelon breeding . In this study, red flesh \extraction method to extract from watermelon flesh carotenoids, compare the advantages and disadvantages of both methods to determine the choice of ultrasonic assisted extraction . Watermelon the extracted carotenoids were separated by column chromatography . Silica gel and magnesium oxide as filler , different concentrations of acetone / petroleum ether mixture as eluent , wherein the acetone content from 0-30% increased stepwise . With red flesh watermelon \Eluate zone of the pigment was measured by UV-Vis absorption spectra of the initial separation of carotenoids identification . The separation and purification by column chromatography on lycopene , β- carotene and ζ- carotene was purified by recrystallization to give a sample of 90% purity , as analyzed by HPLC using standard samples . A column of Vydac 201TP54 C 18 (150 × 4.6 mm); mobile phase A: methanol - dichloromethane - acetonitrile ((7:2:7, V / V), B: methanol gradient elution , 0 min: A: B = 100:0,2 min: A: B = 90:10,4 min: A: B = 80:20,8 min: A: B = 50:50,10 min: A: B = 20:80,15 min: A: B = 0:100,30 min: A: B = 0:100; injection volume : 10μl; column temperature : 25 ℃; flow rate : 1 ml / min ; detection wavelength 400 nm and 450 nm by HPLC conditions , preliminary analysis of carotenoids in watermelon red watermelon flesh mainly carotenoids lycopene and β- carotene -based, and yellow flesh the new watermelon relatively high content of zeaxanthin , other types of carotenoids almost can not be detected , white flesh melon not contain lycopene .
