Abstract


E. coli lipopolysaccharide (LPS), the test several times to stimulate the piglets create stress model, observations proline (PRO) of LPS-stimulated growth performance, plasma biochemical markers, cytokines and intestinal function to explore PRO whether alleviate LPS-stimulated the piglet growth inhibition and intestinal damage. Hybridization select 36 healthy piglets (Duroc × Landrace ×), the average initial weight of 9.13 ± 0.85 kg. Feeding a total of 20 days. Experiment was divided into two phases: the first phase of the trial to 10 days, test the single factor design three dietary treatment groups (control group, 0.5% PRO group and 1.0% PRO group) control diet basal diet, diets of the test group (PRO Group) were added to the basal diet 0.5% PRO and 1.0% PRO. Each treatment group located six replicates, each with two piglets. A piglet separation of the second stage for the trial of 10 to 20 days, respectively, in the three groups of the same repeat reared separately, using a 2 × 3 factorial experimental design, including immune stress (injection of LPS or saline) and PRO added amount (0%, 0.5% or 1.0%), two factors are divided into six treatment groups and each treatment group of six replicates, each repeating a piglet. In test 10, 13 and 20 d of stress treatment group piglets peritoneal injection 100μg/kg · BW LPS, a non-stress control group piglets injected with the appropriate dose of saline. 20 d injection of LPS 2 h at 1 mL / kg · BW dose to all piglets were fed 10% xylose solution. Test the first 10 days and 20 days after injection of LPS 3 hours before vena cava blood sampling, observations relevant indicators. The test observations indicators and results are as follows:. Proline LPS repeatedly stimulate the growth performance and growth levels of hormones and factors detailed records of daily feed intake, testing for 0 d, 10 d and 20 d fasting weighing, calculate the pigs in the first 0 to 10 d, 10 to 20d average daily gain (ADG), average feed amount (ADFI) and feed to gain ratio (F / G), and determination of the two apheresis plasma Epidermal growth factor (EGF), insulin-like growth factor (IGF-1), insulin (INS), glucagon (PG) and somatostatin (SS) level. The results showed that the growth performance of the PRO test 0 to 10 d piglets, dietary supplementation had no effect (P gt; 0.05), but may be to alleviate the trial 10 ~ 20 d LPS multiple stimulus piglet growth performance due to reduced (P lt ; 0.05). Diets supplemented PRO, can improve the first LPS stimulation the piglet plasma EGF level (P lt; 0.05) and multiple LPS stimulate piglet plasma INS (P = 0.08), 0.5% PRO effect better other growth-related hormone has no effect (P gt; 0.05). 2. Proline on LPS repeatedly stimulate piglet plasma biochemical indicators was measured 10 d and 20 d plasma urea nitrogen (BUN), hydrogen peroxide enzyme (CAT), malondialdehyde (MDA), superoxide dismutase (SOD). Results: LPS stimulation of drop increased plasma BUN, CAT, MDA and SOD had no impact (P gt; 0.05); diets supplemented PRO can significantly reduce plasma BUN and MDA, increase plasma SOD (P lt; 0.05) . 3. Proline on LPS multiple stimulus piglets plasma cytokines and stress protein determination of the first 10 days, 3 hours after 20 d injection of LPS piglets plasma interleukin -1 (IL-1), white interleukin -6 (IL-6 ), α1-acid glycoprotein protein (α1-AGP) and heat shock protein -70 (HSP-70). The results show, LPS stimulation had no had no effect on plasma IL-1, IL-6, α1-AGP and HSP-70. The PRO reduce first LPS-stimulated the piglet plasma levels of IL-1 and HSP-70, improved several LPS-stimulated the piglet plasma levels of IL-1 and α1-AGP level (P lt; 0.05). PRO on for the first time and several piglets plasma IL-6 stimulation with LPS and LPS stimulation interaction effect (P lt; 0.05). , PRO alleviate LPS stimulation caused by the stress of piglets, by affecting the generation of cytokines and stress protein diets supplemented with 1% PRO better. Proline several times on LPS stimulation of piglet intestinal function by measuring the content of the 20 d plasma D-xylose, the activity of diamine oxidase (DAO) and section 10 d and 20 d plasma nitric oxide (NO), nitric oxide synthase (NOS) and endothelin (ET) content. The results show that LPS multiple stimulus reduced plasma D-xylose increased plasma DAO (P lt; 0.05); diets supplemented PRO plasma D-xylose (P lt; 0.05) reduced plasma DAO ( P lt; 0.05). First after LPS stimulation, LPS stimulation reduces the plasma of NOS, increased plasma ET (P lt; 0.05); diets supplemented the PRO reduce the LPS-stimulated the piglet plasma ET (P lt; 0.05). Show that diets supplemented the PRO can to some extent alleviate piglet intestinal damage caused by LPS stimulation. Conclusion: diets supplemented the PRO can to some extent alleviate the LPS multiple stimulus piglets caused growth inhibition and intestinal damage, and cytokines as well as the improvement of the antioxidant capacity mitigation LPS stimulation caused by the stress of piglets.
