Abstract


Epigenetic changes in the DNA sequence, heritable gene expression changes. DNA methylation is the epigenetic important part. Existing research shows that the level of DNA methylation in embryonic development in a process of change, and will affect important processes, such as character differentiation during embryonic development. MSAP (methylation sensitive amplified fragment length polymorphism (methylation sensitive amplified polymorphism) the technical analysts chicken during embryonic development, changes in methylation sites, this method can be applied to poultry embryonic development, analysis, been apparent important information for genetic change, and secondly, can for the first time data trend of changes in the degree of methylation of the genome of the domestic chicken embryos, and the degree of methylation differences between tissues and organs during embryonic development in the domestic fowl, can reveal through this process relationship may exist between the dynamic variation of DNA methylation and changes in organ development of this study, in order to further study the changes in DNA methylation or other epigenetic modification affecting economic traits lay the foundation. domestic chicken embryos of different time periods for the trial material extracted genomic DNA using methylation level MSAP its initial detection: a total of 880 sites of methylation found, does not exist outside of the single-stranded and double-stranded closed beta methylation sites 1332 results show CCGG sites outside of the single-chain, the proportion of cytosine methylation sites and is 20.48%, and the inside of the double-stranded CCGG sites cytosine methylation 19.30% to 39.78%, the total proportion of the two, wherein the 3-day period meth the proportion of 32.96% to 38.48%, the methylation ratio of the 7-day period, the proportion of methylation of the 12-day period was 39.45%, the proportion of methylation of the 15-day period was 47.73% As can be seen, the changes in the methylation is with forward to the embryonic development showed an increasing trend in this study different tissues and organs of the domestic chicken embryos to 15 days for the test material, genomic DNA was extracted using methylation-sensitive amplified fragment length polymorphism its methylation level detection: Total found that methylation sites of 1379 and 1553, does not exist outside of the single-stranded and double-stranded closed beta methylation sites. preliminary findings CCGG sites outside of the single-stranded cytosine methylation sites ratio of 23.67 %, the inside of the double-stranded the CCGG-bit point cytosine methylation 23.36%, both the total proportion of 47.03%. pectoral muscle tissue methylation ratio of 47.73%, 42.74% proportion of heart tissue methylation, lung tissue A group the proportion of 39.45% to 46.64%, the methylation ratio of the kidney tissue, liver tissue methylation ratio of 52.48% can be seen, the proportion of liver tissue methylation higher and lower lung, and other relatively close Overall home chicken embryo degree of methylation development presents an increasing trend over time: days of age from 12 days to 15 days methylation relatively high degree of change in the degree of methylation did not change significantly during the 5-12 days. the same time, the domestic chicken embryos methylation presented higher tissue-specific differences, the higher the degree of liver partially methylated, lower lung tissue, and other parts are more close. the embryonic methylation timeline change and embryo weight increase match, embryonic tissue-specific methylation changes also in line with the dynamic process of embryonic organ development and confirmed the the methylation peak period is likely to occur in embryonic form mulberry period, followed by methylation process is still accompanied by embryonic development and change , and there is tissue specific.
