Abstract

Necrotic streaks symptoms field performance for of ToMV, BBWV and CMV monoclonal antibody 315 processing tomato naturally diseased plants ELISA test results show that the ToMV, CMV and BBWV, is the major hazards of processing tomato virus, its composite The infection is causing the symptoms field necrotic streaks main factors. Analysis of serological data Eviews3.0 software found the processing tomato ToMV, CMV and BBWV three viruses accumulation of mutual influence, and positive correlation between each other, a quantitative description of the the different viruses mutual Xiesheng cause The field processing tomato necrosis streak disease seriously endangering the main reason. Resistance Identification by serological testing 21 varieties of field natural incidence tested varieties are not disease-resistant, but the varieties exhibit a certain disease differences. Degenerate primers PA / PB geminivirus Shihezi area performance of 16 processing tomato yellow mosaic and leaf curl symptoms of diseased plants XJ26-1 XJ26-16 PCR, 12 diseased amplified to about 500 bp specificity fragments. Select one representative of diseased plants XJ26-4 PCR products were cloned and sequenced, and the results showed that the fragment of 519 nucleotides (nts), bean golden mosaic virus genus (Begomovirus) DNA-A fragment of the virus genome, and tomato yellow leaf curl virus Y10 isolate (TYLCCNV-Y10, AJ319675) homology of 98.8%. It is suggested that by PCR 500bp fragment of 12 field diseased plants have tomato yellow leaf curl virus infection. According to the measured sequence, the design primers PYF and PYR XJ26-4 DNA-A in the rest of the sequence (2.3kb) cloning, sequencing, sequencing results with 519 nts fragment stitching XJ26-4 DNA-A full-length 2735 nts typical geminivirus genome structural features, and tomato yellow leaf curl virus (TYLCCNV) a high degree of sequence similarity, 91.2 to 99.5%. According to the principle of geminivirus classification, XJ26-4 tomato yellow leaf curl virus XJ26-4 isolates, named Tomato yellow leaf curl China virus-XJ26-4, referred to as TYLCCNV-XJ26-4 English. Monoclonal antibody 23 diseased plants were positive with BBWV2 dsRNA from necrosis streak processing tomato as a template, PCR or RT-PCR using the broad beans virus genus (Fabavirus) degenerate primers Fab5_R1F and Fab5_R1R 19 diseased amplified fragment of the expected size of about 400 bp, select one of the representatives of diseased plants XJ7-5, XJ7-6, XJ7-9 and XJ14-1 PCR products were cloned, sequenced, get XJ7 XJ7-6 and XJ7-9 genomic fragment of 391 NTS, 390 NTS and 392 NTS XJ14-1 of the two positive clones (XJ14-1a the XJ14-1b) of the size respectively 390 nts and 354 NTS, 5 sequence homology between 90.7% to 98.2%. The gene pool to retrieve, they are broad bean wilt virus 2 (BBWV-2) of RNA1 fragments by PCR test, with the the 400bp fragment 19 field diseased plants were BBWV2 to infect. Measured in the sequence of the gene pool the landing BBWV2 of RNA1 nucleotide sequence comparison, four pairs of primers were designed in the RNA1 the sequence conserved region, further sequencing of RNA1 isolates XJ14-1, get XJ14-1 RNA1 RNA2 nucleotide sequence of the gene pool landing BBWV2 sequence alignment, and then according to the the RNA2 sequence conservation area design primer the R2F-1F and R2F-1R. borne, for 93.9%.
