Abstract


Asparagus officinalis is a rich nutritional value of vegetables. The extraction of oligosaccharides isolated from asparagus officinalis is the developing direction of deep processing. The extraction and purification of asparagus officinalis oligosaccharides, and a preliminary identification and function research were studied in this paper. It provided a theoretical basis to further develop the edible and medicinal value of asparagus officinalis.Single-factor test as the basis for four factors and three level orthogonal test, the best conditions of asparagus officinalis raw sugar extracted in hot water is: extraction temperature 80℃, extraction time of 90min, solid to liquid ratio of 1:20, final concentration of 80% ethanol, ethanol precipitation time 8h, extracted 2 times. Using different auxiliary methods to extract asparagus officinalis raw sugar, it shows that: Microwave freeze-thaw Method > Microwave extraction Method > Ultrasonic extraction Method>Flash extraction Method>Hot water extraction. Comparison of sugar content between purple asparagus officinalis and green asparagus officinalis in different parts shows that, the raw sugar in purple asparagus officinalis is about 3.5 times than green ones, and the raw sugar content of the skin was more than the pulp in both asparagus officinalis.The methods of purifying asparagus officinalis oligosaccharides were also tested, the clearance of protein by papain + Sevag method is better than various methods alone. The optimal separation conditions of Sephadex G-25 column were: injection volume 0.4mL, peristaltic pump speed 2r/min, collection volume 1mL.The molecular weight of two asparagus officinalis oligosaccharides were 524.566 and 568.615 determined by GPC. Through IR spectrometry, we find that the form of monosaccharide residues of asparagus officinalis oligosaccharides was a pyran ring or a furan ring in respectively. The composition of asparagus officinalis oligosacch- arides was 1,2,3,4,6-pentakis-O-α-D-Glucopyranose、1,2,3,4,6-pentakis-O- Galacto- pyranose、2,3,4.5,6,-pentakis-pentakis-O-D-Glucose、pentakis-O-Glucopyra-nose and 2,3,4,5,6-pentakis-O-mannosend by analyzing GC / MC spectrometry.With Phen-Fe2+-H2O2 system for the determination asparagus officinalis oligosaccharides scavenging ability, the results show, the scavenging rate of 10mg/mL asparagus officinalis oligosaccharides and 30mg/mL mannitol on hydroxyl radicals was similar. Method of pyrogallol was used to determin the scavenging rate on superoxide anion of asparagus officinalis oligosaccharides, and the clearance rate was 53.30% when the concentration of asparagus officinalis oligosaccharides was 30mg/mL.It shows that asparagus officinalis oligosaccharide is a natural free radical scavengers and antioxidants.In vitro, the GAM liquid medium added asparagus officinalis oligosaccharides can significantly promote the growth of Bifidobacterium, and the differences between the three dose groups were significant (P < 0.05). The proliferation rate of lactobacillus and streptococcus lactis was higher than the control group in both MRS medium added asparagus officinalis oligosaccharides and general poor broth nutrition medium added asparagus officinalis oligosaccharides, but according to statistical analysis, the former and the control group was not significant (P>0.05), the latter with the control group significantly different (P<0.05).
