Abstract


Yersinia pestis as the main object to study the establishment of fiber optic biosensor technology platform, as well as the antigen and antibody detection system, aims to establish on-site rapid and reliable method to detect bioterrorism agent, as well as the realization of pathogens the high sensitivity of detection of antibody. FOB-3 system constitutes completion of the fiber optic biosensor (in cooperation with the Shanghai Institute of Optics and Fine Mechanics). Data processing, system stability assessment and optimization of experimental conditions, the establishment of the FOB-3 detection platform. Yersinia pestis under optimized conditions, the sensitivity of the detection and specific detection analysis and detection of 39 specimens of animal organs. Completion of Yersinia pestis detection analysis on the basis of preliminary detection of Bacillus anthracis spores, detection of staphylococcal enterotoxin (SEB), and yellow fever virus. Established Yersinia pestis F1 antigen detection F1 antibody capture IgG-mediated immunological methods. Expression, purification F1 antigen. By optimizing the experimental conditions, FOB-3 antigens detected best signal value in about 20 min. The sensitivity of the detection protein antigens can be up to 50 to 100 ng / ml (F1 antigen may be detected 50 ng / ml, the SEB the detection to the 100 ng / ml). For multiplication of bacteria can be detected 30-60CFU/ml (EV76 strains of Yersinia pestis 60 CFU / ml, Bacillus anthracis sterne strain 30 CFU / ml), the anthrax the sterne strains of Bacillus detected 4 × 10 CFU / ml. At the same time, the FOB-3 from relatives or far from the source of bacteria in specific identification of the bacteria Yersinia pestis-specific experimental results show. FOB-3 in the actual detection sensitivity 92.6%, specificity 100% of the infected animal samples tested by the Yersinia pestis. The application packet capture FOB-3 can achieve sensitive detection of Yersinia pestis F1 antibody (detected concentration of 10 ng / ml of F1 antibody). Detection of 16 copies of Yersinia pestis infection serum positive signal, sensitivity of 100% and higher sensitivity than the indirect hemagglutination experiments. A specificity of 94.7%, with a total effective rate of 97.1%. FOB-3 pathogenic microorganisms and their structure and metabolites, and Yersinia pestis F1 antibody rapid, sensitive and specific detection. To be further optimized experimental conditions, needs to be improved fiber quality and craftsmanship.
