Abstract

A new study shows, erythropoietin (EPO) is important precipitating factor for proliferative diabetic retinopathy retinal angiogenesis. Tip erythropoietin blockers may be effective in the treatment of this common eye disease. Has proven to vascular endothelial growth factor (VEGF) is the main medium of retinal angiogenesis. However, inhibition of VEGF alone can not prevent retinal angiogenesis, suggesting that other factors also play a role. Known erythropoietin known as pro-angiogenic role, but its role in ocular angiogenesis is unclear. Harvard Medical School, Lloyd Paul Aiello doctors in the comments pointed out that these findings emphasize the erythropoietin, eye disease treatment of patients with diabetes should not only consider the VEGF Attention should also be given to play an important role in the process of changes in visual function factor. The purpose of this study is that the investigation proliferative diabetic retinopathy vitreous humor EPO concentration increased, as well as in animal experiments simulated retinal tissue hypoxia environment and observe the retina in the expression of EPO. First part of proliferative diabetic retinopathy vitreous fluid of patients with erythropoietin hormone concentration was determined Objective To understand the vitreous fluid of patients with proliferative diabetic retinopathy method of change measured by ELISA erythropoietin concentration of 10 normal subjects, 15 patients with PDR patients and patients with 12 patients with vitreous hemorrhage, retinal vein inflammation around the vitreous fluid and serum erythropoietin concentration results 10 normal vitreous fluid concentration of EPO mean 33.5 (19.9-63.8mU/ml) MU / ml, 15 patients patients with PDR glass body fluids, the mean concentration of EPO 880.7 (157.0-1850.0mU/ml,) mU / ml, vitreous fluid of patients with 12 patients with vitreous hemorrhage, retinal vein inflammation around the mean concentration of EPO 396.4 the (83.7-1080.0mU/ml) mU / ml. Significant difference among the groups (P <0.0001). Patients with PDR and vitreous fluid of patients with vitreous hemorrhage, retinal vein inflammation around the EPO concentrations were significantly higher than normal (with vitreous hemorrhage, retinal vein inflammation around the patients vs normal, P = 0.038; the PDR patients vs normal, P < 0.0001). 10 patients with normal serum EPO concentration means 21.3 (16.3-25.5mU/ml) MU / ml, 15 patients with PDR serum EPO concentration in an average of 20.7 (14.4-28.7mU/ml) mU / ml, 12 patients with glass the volume of blood retinal vein inflammation around the mean serum EPO concentration of 25.3 (12.4-63.8mU/ml) mU / ml. No significant difference (P> 0.05) in the group. PDR patients vitreous fluid and serum EPO concentration correlation analysis, vitreous humor EPO and serum EPO concentration and no significant correlation (r = 0.18, P = 0.18). Conclusion PDR vitreous fluid of patients with EPO concentrations were significantly increased, but no correlation with the EPO concentration in the systemic circulation in the blood, suggesting that local lesions caused by diabetic retinopathy concentration. Second part of erythropoietin CoCl 2 -induced rat model of retinal hypoxia express purpose of erythropoietin EPO (erythropoietin, EPO) in the the intravitreal injection COCl 2 expression by chemical-induced rat retinal tissue hypoxia model in rats (body weight 120 ± 20 g) eyes intravitreal injection of CoCl 2 establishment of retinal tissue model of chemical hypoxia, CoCl 2 treatment group rats eye intravitreal injection of 4 microliters 200u mol / l of cobalt chloride, and the other eye injection 4ul of phosphate buffer solution (PBS) as a control, while the normal control group (NC group). After injection respectively 1, 2, 3 rats were killed over the weekend removal of the eye, the use of the immunohistochemistry chemical method to detect the cobalt chloride eyes and normal rat retina prime protein erythropoietin. Results Compared with normal control group, the cobalt chloride treated retinal tissue expression of EPO increased (P <0.05) were significantly different (P <0.01), cobalt chloride group 1, 2, 3 weeks, and tended to increase the expression of EPO with the experimental extension of time, there is a close link between the EPO and the retina local tissue hypoxia injury. Immunohistochemistry method visible expression of EPO retinal cells are mainly located in the ganglion cell layer, inner nuclear layer and outer nuclear layer. Conclusion With the the retinal hypoxia aggravation and time extension of rat retinal expression of EPO increased, and is mainly expressed in the retinal ganglion cell layer, inner nuclear layer and outer nuclear layer.
