Abstract


The anti-inflammatory effects of oxymatrine (Oxymatrine, OMT) has been confirmed by many experiments and has been used in the clinical treatment of hepatitis. But so far for the study of rare lung inflammation reported in the literature. In this paper, the overall level of use of the animals with acute lung injury (Acute lung injury, ALI) model and animal chronic obstructive pulmonary disease (Chronic obstructive pulmonary disease, COPD) model studied OMT intervention effects on lung inflammatory injury. OMT intervention in a rabbit model of ALI in this study with rabbits oleic acid ALI animal model to study the in vivo effects of OMT. The OMT in modeling a dose of 20, 40 and 80 mg / kg. Connected to the polygraph monitor the continuous changes in arterial pressure, heart rate, respiratory rate, blood gas analysis and mining arterial blood sample line tracings. And bronchoalveolar lavage fluid (Bronchoalveolar lavage fluid, BALF), detected in BALF of ICAM-1 and IL-8 concentration. The results found that oxymatrine can significantly reduce the oleic acid model of rabbit lung tissue pathological changes, decreased lung wet / dry weight ratio, increased PaO 2 / FiO 2 The ratio of reduced IL-8 and the content of the ICAM-1 in BALF. OMT intervention in the rat model of COPD study single bleomycin (Bleomycin, BLM) copy tracheal instillation and 27 days of continuous smoked method COPD animal models, research on this platform, the OMT animal models of COPD intervention effect. The OMT starting in the first two days of the BLM instillation continued atomization administered the dose for 200/4mL and 400mg/4mL. After 28 days, the collected blood samples line blood gas analysis, animal spirometer real-time determination of the parameters of lung function in rats. And detecting a concentration of ICAM-1 in the BALF. Oxymatrine significantly reduced rat COPD model group, lung histopathological changes elevated FEV 0.3 / FVC ratio, lower ICAM-1 levels in BALF. The results of this study show that the oxymatrine really has a protective effect on animal lung inflammation model. OMT protective effect on lung injury not only in tissue morphology improvement is also reflected in the functional improvement of lung function and the whole animal. ALI and COPD animal model BALF cytokines oxymatrine, we have reason to speculate that OMT may be generated so as to reach the lung protective effect by inhibiting cytokine excessive.
