Effects of DATS on Acute Lung Injury in Rats

Abstract


Objective To study the diallyl trisulfide right colon ligation and puncture -induced acute lung injury in rats. Methods In this experiment, male SD rats for the study, 30 male SD rats were fasted for 16 hours before surgery , intraperitoneal injection of 10 % chloral hydrate 350mg/Kg anesthesia were randomly divided into three groups (n = 10): control group , CLP group and CLP DATS groups. Control group exposed after laparotomy abdominal intestine, and then layered suture off abdominal surgery after tail vein injection of saline 10ml/kg; CLP group was prepared by cecal ligation and puncture model of acute lung injury , after intravenous injection of 10ml / kg saline ; CLP DATS group cecal ligation and puncture after intravenous injection DATS10mg/kg. All SD rats after subcutaneous injection of saline 5ml/100mg, 12 hours after surgery SD rats were sacrificed specimens of lung tissue samples were measured lung wet / dry weight ratio (W / D), myeloperoxidase ( MPO) activity , malondialdehyde (MDA), superoxide dismutase (SOD) and nitric oxide (NO) content , as well as light microscopy pathological changes of lung tissue . All data were expressed as mean ± standard deviation (x ± s) said, using SPSS11.5 for windows statistical software for data analysis, groups were compared using one-way ANOVA , P lt; 0.05 was considered statistically significant. Results light microscope lung tissue pathological examination showed that the control group alveolar lung tissue did not change significantly under the light microscope , no edema and inflammatory cell infiltration ; compared with control group , CLP rat alveolar wall light microscope edema and thickening , accompanied by inflammatory cell infiltration, alveolar structural damage , focal atelectasis, alveolar exudate appear ; compared with the CLP group , CLP DATS significantly reduced lung injury , but serious than the control group . Compared with control group , CLP group, lung W / D, MPO activity, MDA and NO were significantly increased , SOD activity reduced (P lt; 0.01), while the CLP DATS group compared with the CLP group , lung W / D, MPO activity, MDA and NO content decreased , SOD activity increased (P lt; 0.05). Conclusion DATS can reduce CLP -induced pulmonary edema , inflammatory cell infiltration, reduced MPO activity, MDA and NO content and increased SOD activity in rats with acute lung injury may have a protective effect .
