Abstract


The tiny single-stranded RNA is a class of non-coding RNAs of about 20-25 nucleotides in length , the nematode widely present in the human eukaryotes . miRNA highly conserved , tissue-specific and sequential ; generated miRNA genes are not randomly distributed , from the same precursor RNA processing from the co-expression of miRNA gene cluster exists . The mature miRNA are not completely complementary pairing or fully complementary pairing , causing degradation of the transcription product of the target -specific gene or inducing inhibition of translation , and thus to their 5 ' ends of 5-8 nucleotides , to identify and with target specific gene regulation of the expression of the target gene . miRNA by the above principle in gene regulatory networks play an important role , and also has important implications for research on many aspects of the organism 's ontogeny , cell proliferation , apoptosis , differentiation , and tumor occurrence . The topics, miRNA-125a, miRNA-295, miRNA-181b in mouse preimplantation embryos expression studies , analysis of the possible role of miRNA in preimplantation embryo development, cell differentiation . Objective: To establish superovulated mice , mating and embryo collection system , miRNA -125a , miRNA -295 , miRNA -181b in mouse preimplantation embryos express , explore miRNA in preimplantation embryonic development and cell differentiation the role and significance . Methods: miRNA -specific RT primers for reverse transcription using SYBR Green real-time quantitative PCR studies of miR- 125a , miR -295 and miR-181b expression in mouse preimplantation embryo development process level changes . Results: The various stages of preimplantation embryo development expression of miR-125a, miR-295 and of miR- 181b ; With the development of embryos , the expression of miR -295 is a rising trend in the oocyte ; miR-125a to 2 cell developmental stages of a downward trend , with gradual increase in the future with the development process . Conclusion : miRNA expression in mouse preimplantation embryos ... facing the preimplantation embryo development and differentiation process , which may play a regulatory role .
