Abstract


The gross vein sorrel (Rumex gmelini Turcz.) Polygonaceae Rumex plants. Folk medicine to root, upper respiratory tract infection, gonorrhea, ringworm and sore exact effect, with anti-tumor, anti-fungal, the antitussive asthma, blood pressure, anti-viral and anti-oxidation role. Endophytic fungi (endophytic fungus) is living in healthy plants, fungi of various tissues and organs inside the infected plants do not show obvious external disease, from a strictly surface disinfection plant tissue using histological methods separation method and direct amplification of microbial DNA from plant tissue to prove its endogeneity. R. gmelini endophytic fungi following: (1) R. gmelini endophytic fungi in vitro anti-tumor activity of strains isolated by the screening of this experiment 16 R. gmelini endophytic fungi LOVO, SGC-7901, MCF-7, SMMC-7721, MGC-803, A549, SK-OV-3 in HeLa eight strains of human tumor cells in vitro anti-tumor experiments. MTT assay experiments, record results, using SPSS16.0 software analysis. Screened four anti-tumor effect of endophytic fungi, including bacteria of RGT-S12 on MCF-7, LOVO SK-OV-3 cells with significant inhibition. Bacteria RGT-S31, A549, MGC-803, HeLa cells with significant inhibition. Bacteria of RGT-S42 of SGC-7901, LOVO, SK-OV-3 cells with significant inhibition. Bacteria RGT-S44 has a significant inhibitory effect on SMMC-7721, MGC-803, A549 and HeLa cells. (2) R. gmelini screened endophytic fungi molecular identification by rDNA amplification, sequencing and genetic evolution of the three bacteria are filtered out of the analysis, and combined with the foundation for early morphological identification, results showed that the bacteria RGT -S31 for sickle Alternaria genus Fusarium oxysporum, bacteria the RGT-S42 small Cercospora (Cercosporella) fungi, bacteria for RGT-S44, the sickle are uncultured Fusarium fungi. (3) R. gmelini screened endophytic fungi biological characteristics of four endophytic fungi screened cultural characteristics by changing the different culture conditions, the results show that: the fastest growing of bacteria of RGT-S12 in PDA medium ; most suitable PH value of 6, the optimum temperature of 30 ° C; can make good use of lactose and starch as carbon source, using KNO3 as nitrogen source. Bacteria of RGT-S31 in four medium can better growth; optimum pH value of pH 8, the optimum temperature of 30 ° C; can make good use of maltose as carbon source, using KNO3 as nitrogen source. Bacteria of RGT-S42 in basic medium can better growth in Czapek medium slow growth; optimum pH value pH5, the optimum temperature of 25 ° C; can utilize xylose as a carbon source, peptone and NH4NO3 as nitrogen source. Bacteria RGT-S48 in PDA medium can better growth, slow growth in Czapek medium; optimum growth pH of pH 7, the optimum temperature of 30 ° C; good use fructose as a carbon source, the use of NaNO3 as nitrogen source. This experiment R. gmelini endophytic fungi in vitro anti-tumor experiments, screened four endophytic fungi in vitro anti-tumor effect, on and screening of endophytic fungi were identified, and cultural characteristics of the development of the gross pulse The sorrel endophytic fungi resources and provide the basis to find new anticancer drugs.
