Abstract

It has been reported. This study, animal breeding experiments, the water isocarbophos exposure parental rats were observed Isocarbophos pathology and offspring growth and development of the reproductive function of male rats, and to provide a scientific Learn more about the toxic effects of the Isocarbophos basis. In addition, the combination of the experimental Isocarbophos male rat hypothalamic - pituitary - gonadal axis structure, to explore Isocarbophos, explore new ideas and possible mechanisms of the effects on the male reproductive function. 120 healthy SD rats (80 females, 40 males) were randomly divided into four groups: control group (0mg/kg), low dose group (0.3mg/kg), middle dose group (3mg/kg) high-dose group (6mg/kg). Isocarbophos soluble in cooking oil feed mix, free feeding in rats. After 8 weeks of exposure, female, male 2:1 mated every morning vaginal smear or vaginal plugs Bible Study sperm to be found in the date of the sperm as pregnancy 0D pregnant rats were caged with natural childbirth litter of offspring, the offspring breastfeeding 0,4,7,14 and 21d weigh and measure the length and tail length and chest. After the end of the mating, the male rats were sacrificed, remove the hypothalamus, pituitary, testis, epididymis, prostate and seminal vesicles, weighed and organ coefficient calculated; HE staining and electron microscopy pathological changes; atomic absorption spectrometry determination of testicular tissue calcium, magnesium, copper, zinc content; acquisition rat epididymal sperm, total sperm count and calculate motility, sperm deformity rate. In many ways, to explore Isocarbophos the impact on the structure and function of the male reproductive system in rats. The results showed that: 1 male rats body weight and organ coefficient Isocarbophos the weight gain and organ coefficient of male rats had no significant impact (P GT; 0.05) 2 HE staining (1) the high-dose group hypothalamic neurons visible edema. (2) anterior pituitary eosinophils increased gradually with the increase in exposure dose. (3) after processing Isocarbophos, and the number and level of each dose group testicular tissue spermatogenic cells increased with the exposure dose is gradually reduced; epididymis, prostate, seminal vesicles no significant pathological changes. Electron microscopy (1) increase with the exposure dose increased swelling of mitochondria in the cytoplasm of neurons in the hypothalamus, the high-dose group neurons visible edema; no significant structural changes in glial cells. (2) anterior pituitary growth hormone the perinuclear gap widened, the expansion of the endoplasmic reticulum and mitochondrial swelling in a dose-dependent increase; With the increase of the exposure dose, gradually increased gonadotropin cytoplasmic swelling of mitochondria; adrenocorticotropic hormone cells cells of thyroid-stimulating hormone and prolactin cells showed no obvious structural changes. (3) testicular tissue in the high dose group support cell edema; the spermatogonial cells increase with exposure dose edema until the spermatogonial cell necrosis, membranous structural damage; spermatocytes and sperm cells in each dose group showed no obvious structure change; sperm tail cross-section of the high-dose group \4 testicular tissue calcium, magnesium, copper, zinc content in testis tissue content of calcium, copper dose dependent increase in copper content of the high-dose group compared with the control group difference was statistically significant (P lt ; 0.05); magnesium, zinc content increased with exposure dose and reduce the high dose group compared with the control group were significantly different (P lt; 0.05). 5 sperm count, sperm motility and sperm deformity rate each dose group sperm count, sperm motility than the control group, the difference was statistically significant (P lt; 0.01 and P LT; 0.05); each dose group sperm deformity rate in the control group, the difference was statistically significant (P lt; 0.01 and P LT; 0.05). Reproductive indicators of parental high dose group of rats mating success, survival at birth were lower than the control group, the difference was statistically significant (P lt; 0.05). 7 offspring growth and development of each dose group of offspring body weight, chest circumference, body length, tail length were lower than the control group, a statistically significant difference (P lt; 0.05, P lt; 0.01 or P lt; 0.001); offspring growth and development of each indicators with the exposure dose increased gradually reduced. From the above results can be speculated that Isocarbophos can directly lead to tissue injury of the rat testis seminiferous tubules students at all levels to reduce the number and level of sperm cells, and make spermatogonia, Sertoli cells, spermatogenesis ultrastructural changes , and then decrease in the total number of sperm, reduced sperm motility and sperm deformity rate, which affects the sperm quality of male rats. The Isocarbophos interfere with sperm production by affecting testicular tissue calcium, magnesium, copper, zinc content development and maturation, and ultimately affect the quality of reproductive capacity in rats and offspring. In addition Isocarbophos can lead to the hypothalamus - pituitary - testicular axis pathological structural changes, suggesting that hormone secretion may affect one of the mechanisms of male reproductive function in rats Isocarbophos.
