Abstract

Formaldehyde is a major chemical pollutants in our country every year more than a million workers occupationally exposed to formaldehyde environment, environmental exposure to formaldehyde. Ways to produce formaldehyde contamination variety, such as automobile engine exhaust, smoke release smoke, household materials, such as furniture, particle board, plywood and carpet. International Cancer Research (IMF) classified formaldehyde as a human carcinogen, and that there is a causal relationship between formaldehyde occupational exposure and leukemia, however, strong evidence of this assertion, but insufficient. Therefore, formaldehyde and leukemia relations has become a hot research and debate. Some experts believe that the formaldehyde pollution can induce leukemia, they believe that formaldehyde can induce leukemia through three mechanisms: a direct mechanism of action, namely formaldehyde through a direct effect on the bone marrow cells to induce leukemia, the other two indirect mode, formaldehyde first injury hematopoietic stem cells in the peripheral blood or nasal and oral original pluripotent cells, then these damaged cells into the bone marrow cavity through the blood circulation into bone marrow stem cells to induce leukemia. Some scholars believe that the formaldehyde does not lead to leukemia, reason: formaldehyde has a strong activity into the body, it is easy to degradation in vivo formaldehyde dehydrogenase, and its toxicity can not reach the bone marrow. The toxicity of formaldehyde can reach the bone marrow, that become the focus of debate of the issues the formaldehyde with remote toxicity. In order to investigate whether the formaldehyde with remote toxicity, In this experiment, male Kunming mice as experimental animals, 72h exposed to different concentrations of gaseous formaldehyde, from three levels to study the long-range toxicity of formaldehyde. First, at the cellular level, by measuring the peripheral blood lymphocytes and hepatocytes micronucleus rate to detect damage of the blood cells and cells of the organ of formaldehyde. : Peripheral blood lymphocytes of each exposure group compared with the control group cell micronucleus rate has a very significant difference (P lt; 0.01), liver cell micronucleus rate increases with the formaldehyde concentration increased, indicating that the formaldehyde Heritage injury of peripheral blood lymphocytes and liver cells. Secondly, at the molecular level (DNA level), by measuring the the hepatocyte DNA polymorphism DNA (RAPD) and bone marrow cells polymorphism (RAPD) detecting the toxicity of formaldehyde can reach the liver tissue and the bone marrow cavity, and impact on their genetic material DNA. The results showed that: DNA polymorphism changes of liver cells and bone marrow cells are increased exposure concentration increases, arrived formaldehyde liver tissue, and bone marrow cavity, and their genetic material DNA caused injury. Third, the level of gene transcription (RNA level), regulating the detection of hematopoietic transcription factors HOXB family changes in the level of gene transcription. The results showed that exposure group transcription level compared with the control group showed a downward trend, indicating that formaldehyde can affect the level of regulation of hematopoietic transcription factor regulation.
