Abstract


1 - amino cyclobutoxy , formic acid is an important synthetic amino acids . Currently 1 - amino- cyclobutoxy formate and synthesis methods are mainly a 5 - cyclobutyl - hydantoin ring opening method and cyclobutanone France . Both methods have obvious drawbacks, such as long synthetic route , intermediate compound purification difficulties , high production costs , and is not suitable for industrial production . This article draw glycine synthetic process, the design of a new synthetic route , simple, efficient preparation of 1 - amino - cyclobutoxy formic acid . The method of this paper inverse synthetic analysis of formic acid to give 1 - amino - cyclobutoxy two synthons , and locate two synthon corresponding compound , and then design a new synthetic route : from diethyl malonate and a 3 - dibromopropane starting 1 - bromo cyclobutoxy formic acid is prepared in the four -step reaction . 1 - bromo cyclobutoxy formic acid as the raw materials, and isopropanol as a solvent , hexamine as a catalyst , the reaction with ammonia to prepare a 1 - amino - cyclobutoxy formic acid , and to optimize the preparation of intermediates and the final product process. Also uses 5 - cyclobutyl Hydantoin ureide open loop prepared 1 - amino - cyclobutoxy formic . By IR, NMR and mass spectral characterization of the resulting final product and intermediate .
