Growth of eukaryotic cells is closely related with the changes of nutrients and growth factors in the external environment the whole metabolic regulation of the cell is a highly complex and collaborative process conducted by the regulation of gene expression is the most basic level in the whole process . Ribosome as a synthetic protein factory RNA polymerase Ⅰ, Ⅱ, Ⅲ common coordination transcription , which ribosomal RNA gene transcriptional regulation mechanism has been the hot spot of the research of cell growth and cell cycle regulation . Tral protein yeast cell SAGA and NuA4 chromatin remodeling complex common component of protein , these two chromatin remodeling complex study of the role and function of the yeast cells is more thorough . Studies that the Tral protein may be through NuA4 composite participation ribosomal protein gene, the transcription of ribosomal RNA ; through SAGA composite participation pressure inducible gene expression . This topic is the use of the laboratory has been built the Tral 3355 mutation the Tral 3285 3375 double mutant yeast strains , observed their growth under different growth conditions ; through extraction of RNA by Northern blot analysis of different target genes ( His3 His4 RPS7A, Act1, Scr1) expression , found the Tra1 3355 mutation , Tra1 3285 3375 mutant yeast strain in the ribosomal RNA , HIS3, HIS4 , the transcription of the Act1 affected ; unexpectedly discovered that the accumulation of small RNA Tra1 mutant background . The accumulation of these results will be the next step Tra1 mutations on genome-wide expression microarray analysis provides important information . Also SAGA and NuA4 complex of several components of protein plus protein tag (HA3, Myc13) convenience by co-immunoprecipitation and Western detection Tra1 mutant protein SAGA , NuA4 complex integrity . The results of these experiments will help to better understand the role played Tra1 in the eukaryotic ribosome biogenesis and cell growth regulator .
