Abstract


To explore the IFN-γ on abortion hypothalamus - pituitary - ovarian axis (hypothalamus-pituitary-ovary-axis, HPOA) TNF-α expression, this experiment mifepristone (RU486) model of SD rat abortion by intraperitoneal injection of different doses of IFN-γ, using ultra-sensitive immunohistochemical SP method (streptavidin - peroxidase method), observed the pregnant 13 days (d13) SD rats in group A normal pregnancy, miscarriage Model B group, different doses of IFN-γC, D, E group hypothalamus, pituitary, ovary, uterus TNF-α expression, and to explore low-dose IFN-γ has a protective effect on pregnancy whether its mechanism of action, hoping to at during pregnancy how scientific applications IFN-γ to prevent animals from abortion to provide morphological basis. The main results are as follows: 1. Observed the pregnant rat uterus and bleeding colors, A group of fetal pink, beaded, intrauterine bleeding; B, C, D, E group, there are different degrees of abortion like , B, D, E group embryo Zihei smaller than group A, or uterus was bamboo-like, embryo disappeared, uterine bleeding, vaginal bleeding; C group embryos crimson, some embryos disappear, vaginal bleeding, C pregnancy outcomes than group B, D, E groups. 2. B group, TNF-α expression of positive product nuclei of the hypothalamus, neurohypophysis, pituitary intermediate portion of the follicle, corpus luteum was significantly higher than that in group A, in the anterior pituitary significantly lower than group A, in the ovaries and uterus in the matrix A group expression and no significant difference. Showed that: Compared with normal pregnant rats, miscarriage rat TNF-α expression in HPOA immunoreactive products showed an increasing trend, TNF-α is TH1-type cytokines, cell-mediated immunity and delayed hypersensitivity, the embryo immune injury, which is not conducive to increased expression of pregnancy maintenance. 3 Exogenous intraperitoneal injection 1IU / g IFN-γ after, C rats HPOATNF-α expression of immunoreactive products are as follows: Group A compared with: C group, TNF-α expression of positive products suprachiasmatic nucleus of the hypothalamus , preoptic Jumbo nucleus, periventricular nucleus, arcuate nucleus significantly enhanced in the pituitary, corpus luteum, ovarian stromal and uterine expression decreased significantly; same group B compared, C group HPOATNF-α immunoreactivity was significantly reduced . Show: 50 IU / g IFN-γ can weaken abortion rat HPOA some parts of the expression of TNF-α positive products, may also increase the expression of some other parts of the positive material with 1IU / g IFN-γ, 10 IU / g IFN- γ compared to the weakening effect of the expression of TNF-α is weak, is not conducive to maintaining pregnancy.
