Abstract


Lysosomes play an important role as the only acidic intracellular organelles in the cell life activities , perform the update intracellular biological macromolecules , to digest the physiological function of the cells that engulf foreign bodies as well as processing nutrients . Under normal physiological conditions , lysosomes have good integrity and stability once the lysosomal integrity destruction will not only cause the cell's own physiological features of the disorder , but also has certain cytotoxic , may induce apoptosis or necrosis . Previous studies focused mainly in lysosomal destabilization of cellular effects , but little is known about the pathophysiological factors and mechanisms leading to the loss of lysosomal integrity . Recent studies have shown that significant goods increased lysophosphatidylcholine and phospholipase A2 levels in some cytopathic damage is not yet clear whether these biologically active substances will lysosomal membrane structure . This paper confirms lysophosphatidylcholine and phospholipase A2 can induce lysosomal destabilization , and to clarify the mechanism of lysosomal instability . The free enzyme activity by measuring lysosomal marker enzyme , membrane potential , proton leak , lysosomal internal pH , as well as their latent loss of hypotonic sucrose , we are mainly drawn the following conclusions : lysophosphatidylcholine and phospholipase A2 treatment increased potassium ion permeability of the lysosomal membrane proton permeability , the above-mentioned changes in the lysosomal membrane for potassium ions across the K / H mechanism into lysosomes . In addition , lysoPC PLA2 treatment increases the penetration of the sensitivity of the lysosomes . Transmembrane osmotic pressure imbalance caused by lysosomal penetration instability , is a key factor that led to the loss of integrity of the lysosomal .
