The purpose of vascular injury due to the endodermis integrity faults in the subsequent neointimal hyperplasia process plays an important role. Therefore, the re-endothelialization is a determining factor in inhibiting neointimal hyperplasia. The experimental observation granulocyte colony-stimulating factor (granulocyte-colony stimulating factor, G-CSF) mobilization of circulating CD34 cells accelerate rat injured artery re-endothelialization, thereby inhibiting neointimal hyperplasia effect. Methods SD rats were divided into a treatment group and a control group (n = 20). The treatment group received a subcutaneous injection of recombinant human granulocyte colony-stimulating factor (100μg/kg), 1 times / d, a total of 8d. Subcutaneous injection of normal saline control group. Treatment 5d after intraperitoneal injection of anesthetized rats, the neck along the midline abdominal incision exposed the left common carotid artery 2F arterial embolectomy catheter inserted into the left common carotid artery bifurcation inject 200μL air balloon dilatation, and then back to pull the balloon to at of supraomohyoid, Withdrawing air again returned to the catheter, remove the catheter repeated three times after ligation of the external carotid artery. (1) before treatment and 5d, all rats were removed and 1mL blood leukocytes and CD34 cell count. ② after balloon injury 14 and 28d, each anesthesia, the rats were sacrificed 10, take the left common carotid artery. Each set of the 5 exposed artery intima cavity, the application graphical analysis software calculated re-endothelialization area, area = not Evans blue coloring area / damage total area of ??re-endothelialization; each group and another five, take the left common carotid artery blood extraction cross-section, HE staining using image analysis software neointimal and medial ratio (I / M), to assess the degree of neointimal hyperplasia. ③ detected by immunohistochemistry vascular intima of CD31 of vWF level of cells, to assess the damage vascular reendothelialization. Results ① WBC and CD34 cell: treatment 5d treated rats WBC count was higher than that in control group 2.7 times [(27.60 ± 2.45) are × 10 9 The number of cells is higher than that in control group 12.2 times [(38.31 × 10 7 L -1 , 3.14 × 10 7 L -1 < / sup>, P <0.01)]. ② blood vessels to the extent of re-endothelialization: after balloon injury 14 and 28d, the treatment group re-endothelialization area were (68.3 ± 8.3)%, (97.6 ± 4.1)%, higher than that of the control group [(33.8 ± 6.3)%, ( 76.1 ± 5.2)%, P <0.01]. ③ degree of neointimal hyperplasia: after balloon injury 14,28 d, neointimal and medial of the treatment group were 0.39 ± 0.11, 0.45 ± 0.09, were lower than the control group (0.87 ± 0.15,1.26 ± 0.16, P < 0.01), the degree of neointimal hyperplasia treatment group was significantly suppressed. The the ④ the the vascular intimal of CD31 and vWF cell: after balloon injury 28d, the intima of the treatment group was close to the continuous complete CD31 and vWF cell coverage control group intimal only see sporadic CD31 scattered with vWF cells. Conclusion granulocyte colony-stimulating factor can promote rats after vascular injury in peripheral blood CD34 cell expression and intimal endothelium and inhibit neointimal hyperplasia.
