Objective To study the bone marrow of the mechanisms involved in the pathogenesis of asthma, analysis of mice in the state of sensitization and asthma CD34 positive (CD34 ~) cells in the bone marrow, the expression of CD34 ~ cells in the peripheral blood CD34 ~ cells and lung tissue, and bronchoalveolar lavage fluid (bronchoalveolar lavage fluid, BALF), serum and bone marrow in interleukin-5 (interleukin-5, IL-5) changes in the level, and to observe the role of bone marrow in the pathogenesis of asthma. Method healthy male mice (body weight 18-22g) were randomly divided into three groups of 10. ① asthma group: 10μg ovalbumin (Ovalbumin, OVA) and 10mg aluminum hydroxide by intraperitoneal injection of sensitization to 1% OVA aerosol inhalation stimulate mouse asthma model. (2) sensitized group: be saline (normal saline, NS) inhalation after the sensitization of the above methods. ③ normal control group: NS to the same dose of intraperitoneal injection and inhalation. 24 hours after the last shot harvested lung tissue using HE staining pathological changes and eosinophils (eosinophils, EOS) counts detected in peripheral blood EOS number; flow cytometry of bone marrow and peripheral blood CD34 ~ cell ratio; immunohistochemical method for the determination of the expression of CD34 ~ cells in the lung tissue; ELISA (Enzyme-Linked Immunosorbent Assay, ELISA) to detect the level of IL-5 in BALF, serum and bone marrow. 1 counts and peripheral blood of asthmatic mice lung tissue in EOS EOS proportion higher than those of the normal control group. Compared with normal control mice sensitized mice bone marrow CD34 ~ cell ratio appears higher asthmatic mice further increased. In the the EOS number of peripheral blood of lung tissue EOS ratio, the proportion of bone marrow CD34 ~ cells has a positive correlation. Asthmatic mice peripheral blood of CD34 to cell ratio compared with the sensitized group, the control group increased CD34 ~ cells in their lung tissue expression compared to these two groups increased. Asthmatic mice BALF IL-5 levels than the sensitized group, the control group increased in the BALF of sensitized mice, IL-5, compared with the control group showed no higher. Compared with the normal control group, sensitized mice serum levels of IL-5, and further increased in asthmatic mice. ELISA method can be detected in the sensitized group, asthma group and normal control mice bone marrow IL-5, but no significant difference among the three groups. Conclusions marrow involved in the pathogenesis of asthma. Of hematopoietic progenitor cells in mouse bone marrow in the state of sensitization and asthma have a different degree of proliferation. How much the peripheral blood of lung tissue EOS EOS proportion of bone marrow CD34 ~ cell ratio, positive correlation. 2 mice asthma condition increased expression of CD34 ~ cells in the peripheral blood and lung tissue. Hematopoietic progenitor cells in the bone marrow may asthma outer circumference migrate to the the local airways and lung tissue involved in the formation of the allergic inflammation of asthma. 3. BALF in IL-5 mice were sensitized state did not show increased asthma was significantly higher. The serum IL-5 in the sensitized state i.e. increased, further increase in asthma. The mouse bone marrow in the presence of IL-5, but no obvious change in the state of sensitization and asthma. Compared with the the local airway of IL-5, IL-5 in serum bone marrow involved in the pathogenesis of asthma has played a greater role, but this role may not be decisive, partial bone marrow IL-5 participation mechanisms in the bone marrow in the role deserves further study.
