Abstract


In this study, the different varieties of sweet-scented osmanthus group shoot tips as explants, explore a different medium and different growth regulating substances and other factors in the group shoot tips of each species germination, proliferation and induce rooting process, which aims to filter out for each species group in the different stages of tissue culture medium formulation. Experiment to, Tan Kwai and Kim plantlets leaves as explants, these two sweet-scented osmanthus callus induction, proliferation studies. The trial design uses a single factor randomized block design, orthogonal design and uniform design. The results are as follows: 1. Kim, Yingui, Tan Kwai Meng germination of 0.1% mercuric chloride for disinfection reagents, the appropriate sterilization time range of 3 to 7min 5min. Explants shoot tips as well, compared with the stem segments, disinfection is easy to grasp and the survival rate is high. Kim shoot tip culture medium B5 6-BA2mg / L NAA0.12mg / L, the germination rate of up to 86.1%; silver Gui shoot tip culture medium: B5 B-BA2mg / L of NAA0.02mg / L and germination rate of up to 85%; the Tan Kwai shoot tip culture medium: of B5 or LMC of 6-BA7mg / L, the average strain differentiation rate was 1.73. The germination rate advantage of differences between the varieties group performance the Kim ≥ Yingui,> Tan Kwai> Osmanthusfragrans. Kim and Yingui shoot tips primary culture were similar; Tan Kwai shoot tip culture the callus phenomenon is more serious; almost failed Osmanthusfragrans shoot tips in primary culture. Proliferation culture medium combinations there are differences in the populations of each species. Kim multiplication culture medium B5 6-BA5.0mg / the L NAA0.04mg / L, the proliferation factor of 2.5; B5 6-silver Gui proliferation to the best medium BA7.0mg / L of NAA0 to 0.05 mg / L, the proliferation factor of 2.2; the Tan Kwai proliferation culture medium B5 6-BA8.0mg / L, the proliferation multiple of 2.8. High concentrations of NAA affect the proliferation of three sweet-scented osmanthus. The 4 Osmanthus best rooting medium: 1/285 6-BA0.05mg / L NAA1.5mg / L or 1/2BS B-BA 0.1 mg / L NAA1.5mg / L, the combination can obtain a more good rooting Quality: rooting rate was 100%, with an average root up to 0.5cm, average root number of 2.38. IBA is not the Tan Kwai rooting effective auxin. 5 hardening transplanting to vermiculite and peat (1:1) as well, transplanting survival rate of more than 80%. 6 to the Tan Kwai Kim plantlets leaves callus induction. Tan Kwai callus induction medium: B5 6-BA0.6mg / L 2,4-D0 .6 mg / L sucrose 30g / L agar 5 g / L; Kim callus induction medium: B5 B-BA1.0mg / L 2,4-D0 .6 mg / L sucrose 30 g / L agar 7g / L, osmanthus Plantlets leaf callus in B5 medium and adding a certain amount of 6-BA medium on to be successful induction; the two osmanthus callus proliferation can be very good in the best induction medium.
