Abstract


Enzymatic synthesis of biodiesel raw materials - the production process - green features of the product, enzymatic biodiesel production process between short-chain alcohols, oils and catalysts for the substrate of lipase in a solvent can not be good compatibility , making biodiesel decreased yield, lipase easy inactivation. Candida sp.99-125 laboratory homemade lipase, succinic acid diester sodium (AOT) reverse micelle system, the new method of soybean oil catalyzed synthesis of biodiesel; investigated solvent polarity, AOT concentration, W , 0 (water and surfactant mass ratio), buffer pH, temperature, and other factors that impact on the the lipase catalytic synthesis of biodiesel. The results show that: Candida sp.99-125 lipase-catalyzed, AOT / isooctane reverse micelle system provides more appropriate microenvironment, 11 W 0 , surfactant concentration for 50 mmol / L, temperature of 40 ℃, buffer pH value of 7 AOT / isooctane reverse micelle system, the molar ratio of methanol to oil 3:1, shaking speed 180 r / min, a 12-hour three times Staged methanol, adding 1 mol equivalent of a single batch of the highest conversion rate can reach 90%. Taking into account the transesterification a long time is not conducive to the production of bio-diesel, to further reaction synthetic diesel esterification of palmitic acid and methanol in water / AOT / isooctane reverse micelle system. The test results show that: the water content W 0 for 7, palmitic acid and methanol, the molar ratio of 1 to 2, only one hour, can reach 92% the yield of the methyl palmitate, kinetic studies : Candida sp.99-125 reverse micelle system lipase-catalyzed esterification reaction meets bisubstrate Ping Pong Ping-Pong Bi-Bi mechanism, kinetic parameters for the V max = 3.94 * 10 -8 mol.L -1 . min -1 . mg -1 , K m palmatic acid = 0.4901mol / L, K m methanol = 0.1055mol / L. Cheap Tongzi oil as raw material, using orthogonal experimental design of the the lipase catalytic synthesis of biodiesel Span series reverse micelle system under The results show that: the type of surfactant and methanol stepwise addition reaction results. JMP software design the Jatropha oil fatty acid ethyl esters in AOT reverse micelles system under synthesis experiments, undertaken an evaluation of the impact of various factors, fitting equation the relevant coefficient R 2 is 0.93, indicating that the results have important guiding significance. Experimental results of the reverse micelle system and lipase reuse adopted in the system, fully hydrolyzed fats and oils, and then step-by-step method of substrate was added methanol, to enable the enzyme activity of the lipase maintained 74 hours, the conversion rate reached 83%, The amount of enzyme treatment 0.79g/10mg.
