Abstract


Acidophilic Thiobacillus ferrooxidans (Acidithiobacillus ferrooxidans), is currently the most widely used to study the depth of bioleaching microorganisms. Bacterial adsorption to mineral surfaces both for the survival of the bacteria in the natural environment, or for the bioleaching process has a very important significance. Cell wall-associated protein expression profiling build acidophilic Thiobacillus ferrooxidans bioleaching microorganisms from proteomics angle to explore the surface structure of the study provide some basis for microbial mineral surface adsorption mechanism. This thesis, extracted acidophilic Thiobacillus ferrooxidans cell wall-associated protein liquid chromatography - tandem mass spectrometry technology first digested protein separation, the separated peptides directly into the mass spectrometer ion source a and tandem mass spectrometry analysis. Can get A.ferrooxidans peptide mass fingerprinting (PMF) database are not perfect, we make use of the the bioinformatics technology built for mass spectrometry proteomics analysis A.ferrooxidans PMF database. 245 peptide mass fingerprinting protein information database search using the Mascot software, then the output signal of each protein subcellular localization prediction, currently a total of 97 target proteins. These have been identified proteins 42.3% sec-type signal peptide, 7.2% has a double-arginine transporter signal peptides (Tat), 18.6% of the protein having a lipoprotein signal peptide (Lipo), 21.6% of the protein is a non-classical secretion protein (non-classically secreted proteins), according to the subcellular localization prediction, there are 19.6% of the protein has also been considered a target protein. Classified according to their function, 43.3% of the cell wall protein coated cells, 16.5% protein, 6.2% protein hypothetical protein conservative assumptions protein, 6.2% protein protein of unknown function, other functions of the protein content of small; the molecular weight of these proteins are mainly distributed in 4000 to 118000Da pH value is mainly distributed between 4 to 12, wherein the 31.96% of the protein in pH is less than 7,68.04% of the protein at pH values ??greater than 7,10.31% Protein pH greater than 10.
