Abstract

This study is based on 50-58 weeks old Beijing fatty chicken breeds cock for the test sample, the local species of rooster breeding late on semen quality the best dietary energy and protein levels, and further validation through incubation experiments. For more in-depth study of the molecular mechanisms that affect semen quality, the test for the first time using digital gene expression profiling (DGE) kinds of cock azoospermia testes and fine testicular RNA samples build expression profiling, preliminary analysis of differentially expressed genes . Test the effects of dietary energy and protein levels on the kinds of cock late reproductive performance. The nine treatments were randomly divided into 99 50-week-old oil chicken breeds cock, each handling 11 chickens were fed energy (11.30MJ/kg 11.72 MJ / kg and 12.14 MJ / kg) and protein (12% , 14%, 16%) 3 × 3 9 diets factor combinations. The test results show that the protein levels on growth performance and reproductive performance impact (P gt; 0.05) tends to decrease sperm motility increased with the protein; energy of 12.14 MJ / kg of feed intake and sperm motility significant effects (p lt; 0.05); 11.30MJ/kg-12% group malformation rate was significantly higher than the other groups (P lt; 0.05); stronger interaction between energy and protein, hatching Validation studies have shown 11.72 The fertilization rate MJ/kg-12% better hatchability and child health rates. The Beijing oil chicken breeds cock late dietary protein level of 12%, and the energy level of 11.72 MJ / kg. Test two applications digital gene expression profiling (DGE) to establish the kind of cock azoospermia testicular gene expression profiling, preliminary analysis of the differences in gene expression and function. This test screening rooster testicles one is not producing the kinds of normal sperm kinds of cock and a semen sample, extracted testicular RNA to establish the expression profile analysis of the azoospermia normal kinds of rooster testicles gene expression. The results show that, the of azoospermia testes and normal testis total of 16,379 genes exist differential expression of 545 upregulated genes and 202 downregulated genes. The differentially expressed genes mainly involved in the metabolism of nicotinic acid and nicotinamide, hematopoietic stem cells to produce cells to produce biological pathways of cell cycle and cell structure of the Wnt signaling pathway, and inflammatory response. Spermatogenesis, protein metabolism, mitochondrial structure, the respiratory chain component, and affect a variety of high catalytic activity of the gene expression in normal testis samples, which may be a normal sperm generated reason. Negative regulation of protein kinase testicular azoospermia and inflammation, abnormal metabolism, such as protease inhibitors active gene expression, these factors may lead to sperm production disorders. Establish breeding roosters azoospermia expression spectrum for further analysis of the molecular mechanisms of spermatogenesis and explore the cause of azoospermia would be helpful.
