Abstract

Objective: we use a free-drink model on peripheral neuropathy of chronic alcoholism and discuss the possible mechanism of effects electroacupuncture treatment on CAPN through experimental research, which can provide the new theoretical basis for clinical expansion of the acupuncture treatment of chronic alcoholic peripheral neuropathy.Method: Eighty Wistar rats are divided into two groups randomly: control group and experiment group. After the fourth months,We divided the models into 4 groups randomly. They are model group, methycobal -controlgroup,acupuncture-treated group and electroacupuncture-treated group. We observed the effect of electroacupuncture treatment on weight、pressure pain threshold、sciatic nerve conduction velocity、pathomorphology of sciatic nerve and dorsal rootganglia , NGF and its factor P75mRNA and TrkAmRNA level in sciatic nerve and dorsal rootganglia on model rats by the technic methods of electrophysiology,pathology, immuno-histochemistry,elisa and molecular biology.Results:1、results of making model :(1) The increase of body weight in model group was obviously slower than that in control group.(2)The pressure pain threshold in model group was significantly highter than that in control group after the end of three month (P<0.05).(3) Electrophysiologically the model group rat showed slow motor conduction velocity (MCV) by the end of four month, compaired with control group(P<0.01).(4) HE stain of sciatic nerve of the rat at fouth month end in light microscope showed axonal degeneration accompany with secondary segmental demyelination, but the structure of ganglia showed non apparent abnormality.2、After four weeks treatment: (1) The body weight in all treatment groups was slightly increased than that in model group,there was no statistical significance(p﹥0.05). (2)After treatment of electroacupuncture the pressure pain threshold was decrease slightly than that in model group ,there was statistical significance(p﹤0.05),but the pressure pain threshold in methycobal–control group and in acupuncture-treated group compaired with the model group, there was no statistical significance(p﹥0.05). (3) Electroacupuncture treatment can improve the nerve conductive velocity,reduce the changes of pathological morphology and ultrastructure in sciatic nerve,compared with methycobal-control group and acupuncture -treated group,there was statistical significance(p﹤0.05). (4) Electro- acupuncture treatment can increase the expression of NGF,mRNA of P75 and TrkA in sciatic nerve and DRG. Compared with methycobal-control group and acupuncture -treated group,the difference were significant ( P<0.05) .Conclusion:1、Use a free-drink of increased graduallythe concentration of alcohol can successfully established animal model on peripheral neuropathy of chronic alcoholism.2、Electroacupuncture treatment can improve sciatic nerve conduction velocity of CAPN rats.3、Electroacupuncture treatment can lessen the damages to structure of sciatic nerve and protect the function of sciatic nerve in CAPN rats.4、Electroacupuncture treatment can increase the expression of NGF in sciatic nerve and DRG of CAPN rats.5、Electroacupuncture treatment can promote the expression of P75mRNA and TrkAmRNA in sciatic nerve and DRG of CAPN rats.
