Abstract


The the background matrix metalloproteinases (matrix metalloproteinases, MMPs) are a class of highly conserved proteolytic enzymes can degrade the extracellular matrix, to promote the formation of new blood vessels, thereby promoting tumor cell invasion and metastasis. MMPs can be divided into five major categories, including MMP-2 and MMP-9 is associated with tumor invasion and metastasis of two enzymes. RECK gene is a tumor suppressor gene, to play inhibit malignant tumor invasion and metastasis by inhibiting matrix metalloproteinases. Has been found in the liver, stomach, colorectal cancer, cervical cancer, lung cancer and other tumor tissues RECK, MMP-2 and MMP-9 expression abnormalities. Kidney cancer is a common malignant tumor of the urinary tract. The RECK and MMP-2 and MMP-9 expression in renal cell carcinoma and its correlation has not been reported. The purpose of research RECK, MMP-2 and MMP-9 expression in renal clear cell carcinoma and normal renal tissue, explore the role of RECK and MMP-2, MMP-9 in renal clear cell carcinoma development and their relationship . Experiments using immunohistochemical SP method in 40 cases of renal cell carcinoma and 20 cases of normal kidney tissue were stained to detect RECK, MMP-2 and MMP-9 expression. Results 1.RECK expression in renal cell carcinoma and normal kidney tissue was 55%, 85%, the difference was statistically significant (x2 = 5.275, P = 0.022); of RECK in age lt; 60 years old clear cell renal cell carcinoma tissue and ≥ 60 years of age were positive expression rate was 70%, 52%, the difference no significant sex (Chi-square = 0.175, p 0.676); of RECK in male patients and female patients with renal transparent cell carcinoma tissue The positive expression rates were 48%, 67%, the difference was not statistically significant (x2 = 1.320, P = 0.251); of RECK in the Fuhrman pathological grading of 1 to 2 and 3 to 4 renal clear cell renal clear cell carcinoma cancer tissues, the positive expression rate were 71%, 31%, and the difference was statistically significant (x2 = 6.077, P = 0.014); of RECK in clinical stage Ⅰ ~ Ⅱ and stage Ⅲ ~ Ⅳ The positive expression rates were 67%, 20%, and the difference was statistically significant (x2 = 6.599, P = 0.010). Of MMP-2 expression in renal clear cell carcinoma and normal kidney tissues rates were 73%, 30%, and the difference was statistically significant (x2 = 9.909, P = 0.002); MMP-2 in age lt; positive expression rate of the 60-year-old and ≥ 60-year-old patients with renal transparent cell carcinoma tissue were 82%, 65%, the difference no significant sex (Chi-square = 1.440, p 0.230); MMP-2 in male patients and The positive expression rate in female patients with renal cell carcinoma were 80%, 60%, the difference was not statistically significant (x2 = 1.881, P = 0.170); MMP-2 in the Fuhrman pathological grade 1 to 2 and 3 to 4 renal clear cell carcinoma, the positive expression rate were 58%, 94%, and the difference was statistically significant (x2 = 6.040, P = 0.014); MMP-2 in clinical stage Ⅰ ~ Ⅱ and stage Ⅲ ~ Ⅳ renal clear cell carcinoma, the positive expression rate of 70%, 80%, the difference was not statistically significant (x2 = 0.376, P = 0.540). MMP-9 expression in renal clear cell carcinoma and normal kidney tissues rates were 78%, 25% of the difference was statistically significant (x2 = 15.314, P = 0.000); MMP-9 in the Age lt; 60 years of age and ≥ 60-year-old patients with renal transparent cell carcinoma tissue in the positive expression rate of 82%, 74%, the difference no significant sex (Chi-square = 0.399, p 0.527); MMP-9 in patients with male and female The positive expression rate in patients with renal cell carcinoma were 80%, 73%, the difference was not statistically significant (x2 = 0.239, P = 0.625); MMP-9 in the Fuhrman pathological grade 1 to 2 and 3 to 4 renal clear cell carcinoma, the positive expression rates were 67%, 94%, and the difference was statistically significant (x2 = 4.038, P = 0.044); MMP-9 in the clinical stage Ⅰ ~ Ⅱ and Ⅲ ~ Ⅳ stage renal clear cell carcinoma, the positive expression rates were 73%, 90%, the difference was not statistically significant (x2 = 1.195, P = 0.274). In renal cell carcinoma, RECK and MMP-2 expression was inversely related to the correlation coefficient r = -0.572 (P lt; 0.05); RECK and MMP-9 expression in renal cell carcinoma a negative correlation, the correlation coefficient r = -0.649 (P lt; 0.05). The conclusion 1.RECK showed low expression in renal cell carcinoma and pathological grade and clinical stage, suggesting that RECK act as a disincentive to the development of renal cell carcinoma renal cell carcinoma can be used as a diagnostic indicator and prognosis predictor. Showed high expression of MMP-2 and MMP-9 in renal cell carcinoma and pathological grade, suggesting that MMP-2 and MMP-9 play a catalytic role in the occurrence and development of renal cell carcinoma, can be used as kidney diagnostic indicators and prognostic indicators of clear cell carcinoma. 3.RECK and MMP-2 and MMP-9 expression in renal cell carcinoma showed a negative correlation, suggesting that RECK and MMP-2 and MMP-9 is closely related to impact the occurrence and development of renal cell carcinoma, and RECK renal clear cell carcinoma of the inhibition may be achieved by inhibiting the expression of MMP-2 and MMP-9.
