Background and Purpose : allergic bronchial asthma is a chronic airway inflammatory disease , our children a higher incidence of the disease , and the upward trend . Allergen-specific immunotherapy (allergen specific immunotherapy, SIT) is currently the only known allergic asthma may alter the natural history of the disease due to treatment, but their specific therapeutic mechanism remains unclear. This article by observing dust mite allergic asthma specific immunotherapy (SIT) process, and regulatory T cell function associated transcription factor FOXP3, Th1/Th2 cell differentiation associated with IL-27 p28 and EBI3 mRNA in peripheral blood mononuclear cells (PBMCs) in the expression changes , to study their role in the SIT , and explore SIT treatment of children with allergic asthma pathogenesis , prevention and treatment of asthma, so as to provide new ideas and methods. Methods: According to accept antigen-specific immunotherapy time , collecting the First Affiliated Hospital of Zhengzhou University, November 2007-September 2009 period were treated 39 cases of allergic asthma , its SIT before treatment, after 1 year and treatment two years after the separation of peripheral blood monocytes , extraction of total RNA, reverse transcribed into cDNA, and then use SYBR Green I fluorescence quantitative PCR method to detect each of FOXP3, IL-27 p28 and EBI3 mRNA expression situation . Also, observe the clinical efficacy in children before and after treatment . Results : With the SIT treatment progresses, children with clinical symptoms were relieved , PBMCs of FOXP3 and IL-27 p28 mRNA relative expression levels in specific immunotherapy after 1 year of treatment were significantly higher , respectively, 6.02 times the pre-treatment and 2.18 times ; treatment two years after the FOXP3 mRNA expression decreased for 2.22 times the pre-treatment , IL-27 p28 mRNA expression is also basically recovered pre-treatment levels , is 1.05 times the pre-treatment group ; IL-27 EBI3 mRNA the expression in the healing process as a whole did not change significantly . Conclusion: Effective immunotherapy in children with asthma ; FOXP3 transcription factor and IL-27 in the treatment of asthma SIT process may play an important role .
