Abstract


The elderly due to organ aging, low functional reserve, easily occurs senile lung infections and other incentives under multiple organ dysfunction (MODSE), MODSE lung Start hypothesis. But the lungs start MODSE mechanism is not entirely clear. The genome sequence of the protein will be translated as the function, it is the basic material of organisms to perform various life phenomena. Expression product - protein on the quantity and structure changes observed from the whole genes of a cell or tissue under normal and pathological states, and to help to understand the pathogenesis of the disease, the design-related therapeutic measures. Objective: To establish aged rats with acute lung injury (ALI) model, aged rats ALI lung protein level change for MODSE \Research methods: a small dose of lipopolysaccharide (LPS) intratracheal perfusion method cause acute lung injury model, the contrast observed experimental group and the control group of lung function as well as the changes in pathology and ultrastructural pathology. Using differential proteomics methods, respectively, to the experimental group and the control group protein extract two-dimensional gel electrophoresis. Separation and analysis of gel images to identify differentially expressed protein spots identified by mass spectrometry and biological information retrieval to identify differentially expressed proteins in aged rats ALI. Findings: (1) the experimental group aged rats intratracheally instilled LPS 12hrs, arterial blood gas analysis showed Ⅱ respiratory failure and respiratory acidosis, arterial oxygen pressure (PaO 2 ) oxygen saturation (SaO 2 ) were significantly lower (p <0.05). (2) the experimental group increased rat lung index (0.54 ± 0.04 vs. 0.42 ± 0.02, P <0.01). (3) light microscopy and ultrastructural pathological change of the the LPS group aged rats showed interstitial inflammatory injury. (4) 2-DE gel on the lung tissue of the control group were obtained 730 ± 52, 706 ± 67 points in the the LPS lung tissue on the electrophoresis gel, the ratio was 86%, a doctorate in the People's Liberation Army General Hospital, Postgraduate Medical School the paper Chinese Summary reproducibility better. (5) Through the comparative analysis of two groups gel, to find significant differences in change 16 points. Wherein 13 points abundance LPS group was significantly higher, and there are three points abundance significantly reduces. (6) of these 16 points to cut from the gel, the gel trypsin digestion into peptide fragments, MALDI-TOF-MS analysis to give a clear peptide mass fingerprint peak spectrum (PMF). And eight meaningful proteins after the protein database searches: heat shock protein 27 (heat shock27kna protein): peroxide oxidoreductase (peroxiredoxin); malate dehydrogenase (ma ate dehydrogenase); Valley skin S Gan skin, transferase enzyme (glutathiones transferase); according to the calcium-binding protein I heavy chain (ealpaetin of heavy ehain); the carbonic anhydrase the enzyme (earbon ieanhydraseZ); Rho GDP dissociation inhibitor (Rho GDP the d 1 ssoe at ion inhibitora! pha) ; endoplasmic reticulum protein 29 (endop 1 asm 1 c retue 1 um prote 1 n 29). Conclusion: (l) intratracheal instillation of small doses of LPS induced aging rats acute lung injury; (2) after intratracheal instillation of small doses of LPS induced aging rats with acute lung injury, the pathological changes mainly interstitial inflammation, pulmonary edema and inflammatory cell infiltration interstitial lung; (3) by intratracheal instillation small doses of LPS induced aged rats with acute lung injury, mainly affects the lungs, oxygen exchange capacity, reduced oxygen content in the body, the most famous; (4) doses of LPS induced aging rats with acute lung injury, lung proteome significant abnormal changes in: (5) 2-DE abnormal changes in LPS induced aging rats with acute lung injury lung protein separation; (6) small doses of LPS anomalies are mainly caused by aging acute lung injury, lung proteome response to the stress response to protect cells and involved in inflammation, energy and lipid metabolism, detoxification, protein clear oxidants and free radicals increased expression mainly.
