ISSR Analysis of Genetic Diversity on 21 Lotus(Nelumbo Nucifera) Cultivars

Abstract


Lotus as the only aquatic flower of best ten flowers in China was cultivited for long, which had abundant cultivars, elgent flower appearance and strong fragrance, and it was called for‘ShuiFuRong’by favorite of people all over the world. Lotus was widely cultivated in China, and it played an important role in unban landscape. This paper made a test of genetic diversity and cluser analysis on 21 lotus cultivars by ISSR molecular marker technology, made an classification on some lotus cultivars on molecular level, main results were as follows:1. Optimization of ISSR-PCR reaction system, 25 microlitre reaction system was 2.5μL MgCl2 with a concentration of 5μM/L, 2.5μL 10×buffer, 0.5μL dNTPs of 10mM/L, 1μLprimer of 10μM/L,0.1μL template DNA of 30ng-50ng/μL, 0.5μL taqDNA polymerase of 5U /μL, covering a drop of mineral oil about 30μL, finally added sterile water to 25μL.5. UPGMA cluster analysis method, using this method to make a cluster analysis to 21 lotus cultivars, they were roughly divided into three groups: groupⅠincluded 2 cultivars which were‘Chongbanbayilian’and‘Xiaozuixian’; groupⅡincluded 9 cultivars, which were‘Dajin’,‘Chunhui’,‘Hongbianyudie’,‘Jinzhuluoyupan’,‘Zuilihua’,‘Qiucuilian’,‘Huangying’,‘Gaoweioulian’; groupⅢincluded 9 cultivars, which were‘Kaifengoulian’,‘Liujiangcunoulian’and‘Zhangcunoulian’, ‘Shangqiuoulian’,‘Elianerhao’,‘Baihe’,‘Gulangxiaohong’,‘Baijunzilian’‘Xiaolianzuo’,‘Fenloutai’.
