Abstract

Molecular imprinting technique (MIT) is an emerging interdisciplinary methodology of molecular recognition that is developing in recent years. Molecular imprinted polymers (MIPs) can discriminate template molecule and analogs selectively. Because of their good affinity, sensitivity and stability, MIPs have been rapidly developed and used extensively in biotechnology, clinical medicine, environmental monitoring, inartificial drugs, food industries and so on.Organophosphorus pesticides (OPPs) are a class of pesticides that generally act as cholinesterase inhibitors and are used for the control of a broad range of pests on cotton, rice, tobacco, sorghum, sugarcane and vegetables. The determination of OPPs in soil and water has received considerable attention in recent years. In this dissertation, molecular imprinting polymers were synthesized with monocrotophos as template and applied for analysis of OPPs successfully. The molecular recognition characterstics and binding nature of these MIPs were researched thoroughly by spectrum analysis. The main work of this thesis is summarized as follows:1. Molecular imprinting was used to develop a method based on noncovalent interaction for the synthesis of a monocrotophos-specific polymer. The selective binding characteristics of the imprinted polymer were evaluated by Scatchard analysis and ~1H NMR study. The results focused on the two-point binding nature of the imprinted polymer-monocrotophos interaction and the dissociation constants were estimated to be 30.0μmol/L and 557μmol/L, respectively. Binding study demonstrated that the polymer showed excellent affinity and high selectivity to monocrotophos. The polymer gave more than 15 times longer retention for monocrotophos than the nonimprinted polymer with the same chemical composition.2. A new analytical methodology was developed for the analysis of four polar
