Wheat Transformation of Barley Yellow Dwarf Virus(BYDV) CP Gene via Agrobacterium-Mediated Method

A barley yellow dwarf virus(BYDV)CP gene were transformed on wheat by Agrobacteriummediated method. Two expression vectors were constructed and transferred into five Agrobacterium strains. Nine wheat varieties were used with five kinds of plant acceptors. Resistant plants were obtained. The results of molecular analysis demonstrated that the exogenous gene was integrated into wheat genome. The results are below:1. Wheat mature embryos were obtained and plants were regenerated. The research showed that the raise of inositol concentration is useful for the calli regeneration and ABA addition was useful only for some varieties. Meanwhile, Wheat immature embryos, shoot meristems and anthers were cultured.2. Two plant expression vectors were constructed and transferred into Agrobacterium strains.P18—35sPromoter+CP+nos+NPTIIP19—35sPromoter+EE+CP+nos+NPTIIP18 was transferred into LBA4404(pAL4404), C58 and AGL1;P 19 was transferred into LBA4404(pAL4404), LBA4404(pSB1) and EHA105.3. Immature embryos were transformed. G418 resistant plants were obtained. And the highest frequency was 1.3%.4. The calli from immature embryos were transformed. G418 resistant plants were obtained. and the highest frequency was 2.5%.5. The shoot meristems were transformed. But no plants were positive, according to PCR results.6. The calli from another were transformed. But only obtained G418 resistant calli. no plants were regenerated.7. Resistant plants were planted on soil in green house and bore seeds.8. The results of Southern blotting indicated that the exogenous gene were integrated into wheat genome.
