Abstract


Liver depression is the common TCM symptom-complex. Several prescriptions had been used in CUMS to observe the exchange of behavior and biochemistry matter in hippocampus of rats which are important to the regeneration of neuron. And to find how do TCM treat on the Liver depression. The First Part TCM Characters of the CUMS models1 The effects of Si-ni Powder, Xiao-yao Powder, Si-junzi Decoction,Gui-pi Decoction and Wen-dan Decoction on chronic unpredictable mild stress model behavior changesTo study the effects of Si-ni Powder, Xiao-yao Powder and Si-junzi Decoction ,Gui-pi Decoction and Wen-dan Decoction on chronic unpredictable mild stress model behavior changes. We found that rats exposed to CUMS showed decreased sucrose preference and increased immobile time in forced swimming test, increased content of serum corticosterone. It proved that rats exposed to CUMS appears lower appetition on happinese and inactivity on escaping from dangers, which seems like typical symptoms of liver depression. Si-ni Powder and Xiao-yao Powder prevented theses changes, while Si-junzi Decoction , Gui-pi Decoction and Wen-dan Decoction had no effects. While rats weights did not show obvious difference.2 The effects of Si-ni Powder, Xiao-yao Powder, Si-junzi Decoction,Gui-pi Decoction and Wen-dan Decoction on chronic unpredictable mild stress model behavior changes in different timesTo study the effects of Si-ni Powder, Xiao-yao Powder and Si-junzi Decoction ,Gui-pi Decoction and Wen-dan Decoction on chronic unpredictable mild stress model behavior changes in different times . In the 7th day, rats exposed to CUMS didn’t showed obvious increase content of serum corticosterone and decrease sucrose preference and increased immobile time in forced swimming test. In the 14th day, rats exposed to CUMS didn’t showed obvious decreased sucrose preference and increased immobile time in forced swimming test but showed increased content of serum corticosterone. Si-ni Powder prevented theses changes, and Si-junzi Decoction, Xiao-yao Powder, Gui-pi Decoction and Wen-dan Decoction had no effects. In the 21th day, rats exposed to CUMS showed decreased sucrose preference and increased immobile time in forced swimming test, increased content of serum corticosterone. Si-ni Powder and Xiao-yao Powder prevented theses changes, but Si-junzi Decoction, Gui-pi Decoction and Wen-dan Decoction had no effects. In the 14th day, rats exposed to CUMS didn’t showed obvious changes in expression of 5-HT1AR while showed decreased expression of CREB、BDNF mRNA and protein. Si-ni Powder and Xiao-yao Powder protected theses changes, and Si-junzi Decoction,Gui-pi Decoction and Wen-dan Decoction had no effects. In the 21th day, rats exposed to CUMS showed decreased expression of 5-HT1AR、CREB、BDNF mRNA and protein. Si-ni Powder and Xiao-yao Powder prevented theses changes, and Si-junzi Decoction , Gui-pi Decoction and Wen-dan Decoction had no effects.Then we draw the conclusion that the animal model we used in this study can be named“chronic stress liver depression model”. The change of behavior followed the change of time and associated with the expression of 5-HT1AR、CREB、BDNF mRNA and protein in hippocampus.
