Safety Assessment of Genetically Modified Rice with Bar Gene on Musculus Feeding

Abstract


Molecular biology methods were used in this study for the residue detection of the foreign gene and the related expressed protein in Bar68-1GM rice. Digestion stability of the foreign protein and genetic mutation of mtDNA from small intestine were also studied.(1)60KM mice weigh between18and24g were separated into two groups:the test group and the control one, both of which were raised by the Configured feed. After90days, samples were selected randomly for the foreign gene test by PCR respectively in the mice liver, kidney, spleen, small intestine and crureus. There was no positive results in all above tests, which means the foreign gene wasn’t remained or transferred.(2) The residue of Phosphorus chloramphenicol acetyltransferase (PAT), the expression product of Bar gene in mice organs and digestive system were tested by Enzyme-linked immunosorbent assay (ELISA). There was no color reaction in any well of the microtiter plates (except stomach). This result indicated that PAT could be digested and degraded completly in the body.(3) Refer to the relevant provisions of the Ministry of Agriculture, total protein from both of Bar GM rice and the normal rice were extracted for the use of the vitro digestion stability experiment. Neither the Bar68-1protein nor the D68one could stay stable in the simulated gastrointestinal digestive juice, from which a conclusion could be arrived that PAT does not have the property of digestion stability.(4) Four pairs of oligonucleotide primers were designed for the12s-16s conservated region of the mice intestinal mitochondrial genomic DNA(mtDNA), the PCR amplicons were send to Shanghai Sangon Biological Engineering Co., Ltd. for both of positive and negative sequential testing. A result that no mutational site was found came out after the sequence comparison in NCBI. It did not cause mutations in12s-16s conservated region of mtDNA through the feeding with Bar GM rice.Conclusion:In this experimental condition, genetically modified herbicide tolerant rice with Bar gene had no impact on the structure and function of the musculus digestive system.
