The Dictamnine, bergamot, ursolic was the main active medicinal ingredients in Zanthoxylum dissitum, which have a significant effect on Hypertension, high cholesterol, sore throat and other diseases. In terms of anti-tumor, anti-inflammatory, anti-HIV, anti-diabetic and immunoregulation had been showing good pharmacolog-ical prop-erties. In this paper, optimizing the chromatographic conditions, determine-eing the main active ingredients contents of the Dictamnine and bergamot and Ursolic in the Dictamnine, and ascertain the optimum extraction method. Horizontal comparison different parts and different sources of the active ingredient content in Zanthoxylum dissitum.A high performance liquid chromatography analysis method was established for the active ingredient content in the Zanthoxylum dissitum, choose the most suitable chromatographic conditions of the system:the Dictamnine and bergamot’s mobile phase was acetonitrile/water=4/6(V/V)+0.1%TFA, the column temperature is25℃, flow rate at1.OmL/min, detection wavelength was254nm; ursolic’s mobile phase was acetonitrile+0.1%TFA, the column temperature was25℃, flow rate at1.OmL/min, detection wavelength was210nm; basically reached the baseline and have a good peak shape, and investigated the precision and stability of the instrument for proving accurate and reliable of analytical methods.The process of extraction for mainly constitutents from Zanthoxylum dissitum was tested. Compared extraction methods such as acetic acid, ethanol-water, chloroform, water and reflux, cold soak, ultrasonic for to extracting the dictamnine, bergamot, ursolic. We choose the most suitable conditions of the extraction system: extraction solvent was80%ethanol, ultrasonic extracted30min, extraction temperature was24℃(room temperature).Extracted the active ingredients of roots, stems, leaves in Zanthoxylum dissitum with optimal conditions, detected the difference of the active ingredient content in different parts of the Zanthoxylum dissitum. The experimental results show that the Zanthoxylum dissitum’s root could be extracted the highest active ingredient content, the dictamnine was0.47mg/g, the bergamot was0.35mg/g, the ursolic was0.25mg/g;the lower active ingredient content in stems, the dictamnine was0.39mg/g, the bergamot was0.31mg/g, the ursolic was0.16mg/g; the minimum active ingredient content in leaves, the dictamnine was0.14mg/g, the bergamot was0.09mg/g, the ursolic was0.04mg/g. All the parts of the plant can be used as raw materials. And compared active ingredient content in Zanthoxylum dissitum from four provinces, such as Hunan, Hubei, Gansu, Sichuan. The results show that the average content of active ingredients of different Zanthoxylum dissitum samples are small differences. The highest concentrations were found in Zhangjiajie, the dictamnine was0.49mg/g, the bergamot was0.38mg/g, the ursolic was0.26mg/g, and Xiantao area was the lowest, the dictamnine was0.42mg/g, the bergamot was0.32mg/g, the ursolic was0.23mg/g. The different of the active ingredient content is about10%. This may be due to the role of genes and environment, it lead Zanthoxylum dissitum to gradually form different of growth and development, caused unique active ingredients.
