Abstract


Objective:By Monosialotetrahexosylganglioside(GM1) on the intervention of ExperimentalAutoimmune Encephalomyelitis(EAE) in rats to observe the corresponding changesin the expression of CXC chemokine ligand1(CXCL1) and Neurofilament protein(NF)in the spinal cord,understanding whether GM1have a protective effect for MS and itsmechanism,and thus for clinical treatment of MS to provide new ideas andexperimental evidence.Methods:A total of72adult healthy female Wistar rats were randomly divided into4groups:normal control group,EAE group,EAE group plus large-dose of GM1group,EAE group plus low-dose of GM1group(n=18).Normal group weresubcutaneously injected by saline water.The EAE model were established withself-made complete antigen,then the large(100mg/kg.d)and low(40mg/kg.d)doses ofGM1were given according the experimental protocol.The incidence and the scores ofneurologic impairment were recorded,also the Bielschowsky silver staining,thechemokine CXCL1(CXCL1)and neurofilament protein(NF) immunohistochemistrystaining were performed and analyzed.Results:1. The incidence in different groupNone ill rats in the normal control group;In EAE group,the incidence is83.33%;The incidence became lower after the treatment of GM1;The incidence oflarge-does GM1group was lower than low-does GM1group.2. Neural function assessment in different groupClinical signs of EAE were assessed and scores were assigned in the seventh day,the14th day and21th day,the score of GM1group was lower than EAE group inthe14th day and21th day;the score of large-does GM1group was lower thanlow-does GM1group in the14th day;when the three time points were compared,thedifferences of scores with groups in the seventh day and the21th day werestatistically significant when compared with the groups of the14thday(P<0.05).Scores were not significant between groups of the seventh day and the21th day(P>0.05).3.Histopathology3.1Bielschowsky silver stainingIn the normal control group,the vast majority of axonal thicknessuniformity,course direction of a more consistent,the continuity of axonalintegrity.Varying degrees axonal changes appeared in the other groups.Axon appearedswelling,uneven thickness,deformed,pale staining vacuoles,fracture,retraction ballchange.4.Immunohistochemistry4.1Expression of CXCL1The other three groups all have CXCL1expressed except the normalgroup,comparison in each group the number of CXCL1positive cells:CXCL1was notexpressed in the normal group;In EAE group,the expression of CXCL1was most,byGM1intervention,CXCL1expression decreased,and in large-does GM1group,theexpression of CXCL1was significantly decreased than low-does GM1group(P<0.05).From the three time points for comparison,the number of CXCL1positive cells in large-does group were more than low-does group in the14th day;thepositive expression of CXCL1was markedly different between groups of the14th dayand groups of the seventh day and the21th day(P<0.05).the number of CXCL1positive cells were not significant between groups of the seventh day and the21thday(P>0.05).4.2Expression of NFFour groups all have NF expressed,comparison in each group the number of NFpositive cells:In normal group,the expression of NF was most;expression of NF significantly decreased in EAE Group when compared at the same time with normalgroup,by GM1intervention,NF expression increased,and the expression of NF wassignificantly increased in large-dose GM1group than low-does GM1group(P<0.05).From the three time points for comparison,the number of NF positivecells in large-does group were more than low-does group in the14th day;the positiveexpression of NF was markedly different between groups of the14th day and groupsof the seventh day and the21th day(P<0.05).the number of NF positive cells were notsignificant between groups of the seventh day and the21th day(P>0.05).Conclusion:1.The expression of CXCL1and NF in spinal cord are associated with the activity ofEAE rats,GM1intervention on various indicators have an impact.2.GM1have protective effect on EAE in rats by protecting the rats from the severityof the disease,while its effect and dose related.
